Abstract
Aterosklerosis merupakan kondisi terjadinya penyempitan pembuluh darah akibat adanya penempelan plak-plak lemak. Terjadinya penempelan lemak ini salah satunya disebabkan karena terjadinya oksidasi LDL sehingga timbul plak yang akan mudah menempel di pembuluh darah, selain itu juga karena makanan yang dikonsumsi kaya kolesterol. Sambiloto (Andrographis paniculata (Burm.f) Ness) merupakan tanaman obat yang memiliki khasiat menurunkan kadar lemak darah secara praklinik pada tikus. Andrografolid (diterpen lakton) merupakan salah satu senyawa yang terkandung dalam sambiloto yang diketahui bertanggung jawab terhadap aktivitas sebagai antiaterosklerosis. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pemberian isolat andrografolid dalam memperbaiki profil histopatologi aorta hewan uji. Berdasarkan hasil yang diperoleh terdapat perbedaan antara kelompok dosis 3 dengan dosis 30 mg/KgBB dengan kelompok pembanding simvastatin. Maka dapat disimpulkan isolat andrografolid memiliki efektivitas dalam menurunkan kolesterol dan memperbaiki kerusakan aorta lebih baik dari kelompok pembanding simvastatin. --- Atherosclerosis is a condition of narrowing of blood vessels due to the attachment of fatty plaques. One of the occurrences of fat attachment is caused by LDL oxidation so that plaque arises that will easily stick to blood vessels, besides that also because the food consumed is rich in cholesterol. Sambiloto (Andrographis paniculata (Burm.f) Ness) is a medicinal plant that has the property of reducing blood fat levels preclinically in rats. Andrographolide (diterpene lactone) is one of the compounds contained in sambiloto which is known to be responsible for its activity as an antiatherosclerosis. The purpose of this study was to determine the effect of andrographolide isolate administration in improving the histopathological profile of the aorta of test animals. Based on the results obtained, there was a difference between dose group 3 with a dose of 30 mg / KgBB with the comparison group simvastatin. So it can be concluded that andrographolide isolate has effectiveness in lowering cholesterol and repairing aortic damage better than the comparison group of simvastatin.
