Abstract
Singawalang (Petiveria alliance L.) adalah salah satu jenis tumbuhan yang banyak ditemukan di Indonesia, berasal dari hutan hujan Amazon. Terutama pada daunnya yang memiliki manfaat baik bagi tubuh dan mengandung antioksidan. Antioksidan memberikan keuntungan bagi kesehatan, yaitu berperan dalam melawan radikal bebas yang berbahaya bagi tubuh. Metode pengukuran antioksidan dengan menggunakan metode penangkalan radikal bebas DPPH (2,2-diphenyl-1- picrylhydrazl). DPPH merupakan metode pengujian aktivitas antioksidan yang didasarkan pada suatu zat yang mampu meredam radikal bebas. Tujuan dari penelitian untuk mengetahui aktivitas antioksidan dari ekstrak dan fraksi daun singawalang terhadap DPPH dan untuk mengetahui aktivitas antioksidan terbaik dari ekstrak atau fraksi daun singawalang menggunakan metode DPPH. Digunakan vitamin C sebagai pembanding. Hasil penelitian menunjukkan bahwa ekstrak dan fraksi daun singawalang memiliki aktivitas antioksidan terhadap DPPH, aktivitas antioksidan pada ekstrak etanol, fraksi air dan fraksi etil asetat daun singawalang memiliki nilai IC50 berturut – turut 3,371 ;2,785 dan 52,456 ppm. Aktivitas antioksidan terbaik dimiliki oleh fraksi air daun singawalang dengan nilai IC50 2,785 ppm. --- Singawalang (Petiveria alliance L.) is a type of plant that is commonly found in Indonesia, originating from the Amazon rainforest. It's particularly known for its leaves, which offer various benefits to the body and are rich in antioxidants. These antioxidants play a vital role in promoting health by combatting harmful free radicals in the body. The method used to measure these antioxidants involves the DPPH (2,2-diphenyl-1-picrylhydrazyl) free radical scavenging technique. Essentially, DPPH is a testing method that evaluates the ability of a substance to neutralize free radicals. The primary goal of this research was to assess the antioxidant capabilities of Singawalang leaf extracts and fractions when exposed to DPPH. Additionally, the aim was to identify the most potent antioxidant activity among these extracts and fractions using the DPPH method, with vitamin C serving as a reference point. The research findings revealed that both the extract and fractions of Singawalang leaves demonstrated considerable antioxidant activity against DPPH. Specifically, the antioxidant activities in the ethanol extract, water fraction, and ethyl acetate fraction of Singawalang leaves were measured by their respective IC50 values: 3.371, 2.785, and 52.456 ppm. The water fraction of Singawalang leaves exhibited the most robust antioxidant activity, boasting an IC50value of 2.785 ppm.
