Abstract

Andrografolid merupakan komponen bioaktif paling utama yang terkandung dalam tanaman sambiloto (Andrographis paniculata Burm. F. Ness ). Penelitian ini bertujuan untuk mengoptimasi pelarut, pH dan waktu kontak pada proses pemurnian ekstrak sambiloto. Penelitian dilakukan pada pH 2 dan 12, pelarut yang digunakan adalah metanol, etanol, dan etil asetat. Sedangkan waktu kontak yang diteliti yaitu 30, 60, 90, dan 120 menit. Isolat hasil pemurnian dikarakterisasi menggunakan kromatografi lapis tipis (KLT), spektrofotometri UV – Vis, dan kromatografi cair kinerja tinggi (KCKT). Meskipun tidak terjadi pengurangan jumlah noda pada KLT dan pengurangan jumlah puncak pada spektrofotometri UV - Vis, berdasarkan hasil optimasi pelarut, pH, dan waktu kontak didapatkan hasil bahwa pelarut metanol dengan waktu kontak 90 menit, dan pH 2 memberikan peningkatan % area yang paling tinggi yaitu dari 74 % menjadi 76,82%. --- Andrographolide is the principal bioactive component found in the Andrographis paniculata Burm. F. Ness plant. This study aims to optimize solvents, pH, and contact time in the purification process Andrographis extract. The research was conducted at pH 2 and 12, using solvents methanol, ethanol, and ethyl acetate. Meanwhile, the investigated contact times were 30, 60, 90, and 120 minutes. The isolated purification product was characterized using thin-layer chromatography (TLC), UV-Vis spectrophotometry, and high-performance liquid chromatography (HPLC). Although there was no reduction in the number of spots in TLC and a decrease in peak numbers in UV-Vis spectrophotometry, based on the results of solvent, pH, and contact time optimization, it was found that methanol solvent with a contact time of 90 minutes and pH 2 yielded the highest increase in the % area, from 74% to 76.82%.
