Abstract

Radikal bebas merupakan senyawa reaktif yang dapat memicu reaksi berbahaya bagi tubuh dan menyebabkan penyakit degeneratif. Senyawa antioksidan berperan penting dalam menetralisir radikal bebas dan mencegah kerusakan oksidatif karena mampu menghambat proses oksidasi dengan mendonorkan elektron pada radikal bebas. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan ekstrak air daun sukun (Artocarpus altilis (Parkinson) Fosberg), bunga rosela (Hibiscus sabdariffa L.), dan kombinasinya menggunakan metode DPPH (2,2-difenil-1- pikrilhidrazil) berbasis microplate reader. Proses ekstraksi dilakukan dengan metode dekoktasi, kemudian dilakukan pengujian DPPH terhadap masing-masing ekstrak dengan variasi konsentrasi bertingkat. Hasil penelitian menunjukkan bahwa nilai IC₅₀ asam askorbat sebagai pembanding adalah 7,698 µg/mL, yang menunjukkan aktivitas antioksidan sangat kuat. Ekstrak daun sukun memiliki nilai IC₅₀ sebesar 779,083 µg/mL, ekstrak bunga rosela memiliki nilai IC₅₀ sebesar 625,329 µg/mL, dan kombinasi ekstrak daun sukun dan bunga rosela 2:1 memiliki nilai IC₅₀ sebesar 754,628 µg/mL. Berdasarkan nilai IC₅₀, ekstrak air bunga rosela menunjukkan potensi paling tinggi diantara ekstrak lainnya. Kombinasi ekstrak daun sukun dan bunga rosela pada rasio 2:1 lebih lemah dari yang diharapkan dan tidak menunjukkan efek sinergis karena tidak meningkatkan efektivitas antioksidan rosela tunggal. ------ Free radicals are reactive compounds that can trigger harmful reactions to the body and cause degenerative diseases. Antioxidant compounds play an important role in neutralizing free radicals and preventing oxidative damage because they are able to inhibit the oxidation process by donating electrons to free radicals. This study aims to determine the antioxidant activity of breadfruit leaf water extract (Artocarpus altilis (Parkinson) Fosberg), rosela flower (Hibiscus sabdariffa L.), and its combination using the DPPH (2,2-diphenyl-1-picrylhydrazyl) method based on a microplate reader. The extraction process is carried out by the decocation method, then DPPH testing is carried out on each extract with multi-level concentration variations. The results showed that the IC₅₀ value of ascorbic acid as a comparison was 7.698 μg/mL, which indicates very strong antioxidant activity. Breadfruit leaf extract had an IC₅₀ value of 779.083 μg/mL, rosela flower extract had an IC₅₀ value of 625.329 μg/mL, and the combination of breadfruit leaf extract and rosela flower 2:1 had an IC₅₀ value of 754.628 μg/mL. Based on the IC₅₀ value, rosela flower water extract shows the highest potency among other extracts. The combination of breadfruit leaf and rosela flower extract at a ratio of 2:1 was weaker than expected and did not show a synergistic effect as it did not increase the antioxidant effectiveness of a single rosela.
