Abstract

Herba Rumput Mutiara (Oldelandiae corymbosa L.) mengandung senyawa aktif seperti fenolik, flavonoid, tanin, terpenoid, dan saponin yang berpotensi sebagai antioksidan alami. Penelitian ini bertujuan untuk mengidentifikasi, dan mengisolasi kandungan senyawa bioaktif dalam fraksi air dari ekstrak etanol herba rumput mutiara (Oldelandiae corymbosa) yang memiliki aktivitas antioksidan. Fraksinasi dilakukan menggunakan metode ekstraksi cair-cair dengan pelarut n-heksan, etil asetat, dan air. Fraksi air dipilih karena menghasilkan rendemen tertinggi sebesar 59,8037%. Identifikasi aktivitas antioksidan dilakukan menggunakan metode kromatografi Lapis Tipis (KLT) dengan penambahan pereaksi DPPH (2,2-diphenil-1-pikrihidrazil) dan fase gerak kloroform : metanol (3:1). Subfraksinasi menggunakan kromatografi kolom menghasilkan 62 subfraksi, dan vial 11-15 yang digabung dari kolom ke-2 terpilih untuk proses isolasi. Isolat dimurnikan dengan KLT preparatif, diuji kemurniannya dengan KLT dua dimensi menggunakan eluen etil : metanol (2:4) dan kloroform : etil (7:3). Hasil analisis menunjukkan bahwa isolat merupakan senyawa golongan fenolik dengan aktivitas antioksidan, ditandai oleh spektrum serapan khas gugus -OH dan perubahan warna saat uji pereaksi geser serta respon positif terhadap DPPH. ------ Pearl Grass Herb (Oldenlandiae corymbosa L.) contains active compounds such as phenolics, flavonoids, tannins, terpenoids, and saponins that have potential as natural antioxidants. This study aimed to identify and isolate bioactive compounds in the aqueous fraction from ethanol extract of pearl grass herb (Oldenlandiae corymbosa) that possess antioxidant activity. Fractionation was performed using liquid-liquid extraction method with n-hexane, ethyl acetate, and water as solvents. The aqueous fraction was selected because it yielded the highest recovery of 59.8037%. Identification of antioxidant activity was conducted using Thin Layer Chromatography (TLC) method with the addition of DPPH (2,2- diphenyl-1-picrylhydrazyl) reagent and mobile phase of chloroform : methanol (3:1). Subfractionation using column chromatography produced 62 subfractions, and vials 11-15 combined from the second column were selected for the isolation process. The isolate was purified using preparative TLC, and its purity was tested using two-dimensional TLC with eluents ethyl acetate : methanol (2:4) and chloroform : ethyl acetate (7:3). Analysis results showed that the isolate was a phenolic compound with antioxidant activity, characterized by specific absorption spectrum of -OH groups and color changes during shift reagent test as well as positive response to DPPH.
