Abstract

Kurkumin dikenal sebagai senyawa bioaktif dengan beragam aktivitas farmakologis. Namun, penggunaannya masih terbatas akibat kelarutannya yang sangat rendah dalam air. Penelitian ini bertujuan untuk mengkarakterisasi dan mengevaluasi efektivitas karagenan sebagai polimer hidrofilik dalam pembentukan sistem dispersi padat untuk meningkatkan kelarutan kurkumin. Dispersi padat dibuat menggunakan metode penguapan pelarut dengan variasi rasio kurkumin dan karagenan 1:1, 1:2, dan 1:3. Hasil uji densitas sejati menunjukkan peningkatan nilai densitas dari 1,189 g/mL pada kurkumin menjadi 1,251 g/mL, 1,347 g/mL, dan 1,394 g/mL berturut-turut untuk rasio 1:1, 1:2, dan 1:3. Uji kelarutan menunjukkan peningkatan, dari 2,01% pada kurkumin menjadi 13,42%, 17,78%, dan 30,01%. Pada profil disolusi, nilai puncak tercapai pada menit ke-45 dengan disolusi masing-masing sebesar 2,376%, 3,043%, dan 4,765%. Spektrum FTIR menunjukkan tidak adanya pembentukan senyawa baru pada sistem dispersi padat. Analisis SEM menunjukkan perubahan morfologi menjadi bentuk globular yang tersebar merata dengan permukaan lebih halus. Berdasarkan hasil tersebut, dapat disimpulkan bahwa karagenan terbukti efektif dalam meningkatkan kelarutan, profil disolusi serta memodifikasi sifat fisikokimia kurkumin melalui pembentukan sistem dispersi padat. ------- Curcumin was recognized as a bioactive compound with various pharmacological activities. However, its application remained limited due to its low water solubility. This study aimed to characterize and evaluate the effectiveness of carrageenan as a hydrophilic polymer in the development of a solid dispersion system to enhance the solubility of curcumin. Solid dispersions were prepared using the solvent evaporation method with curcumin-to-carrageenan ratios of 1:1, 1:2, and 1:3. The true density test results showed an increase in density values from 1.189 g/mL for pure curcumin to 1.251 g/mL, 1.347 g/mL, and 1.394 g/mL for the 1:1, 1:2, and 1:3 ratios, respectively. Solubility tests indicated a enhancement, from 2.01% for curcumin to 13.42%, 17.78%, and 30.01%. The dissolution profile showed peak values at the 45th minute with dissolution rates of 2.28%, 3.04%, and 4.77%, respectively. FTIR spectra confirmed the absence of new compound formation in the solid dispersion system. SEM analysis revealed morphological changes into evenly distributed globular structures with smoother surfaces. Based on these findings, it was concluded that carrageenan was effective in improving the solubility, dissolution profile, and physicochemical properties of curcumin through the formation of a solid dispersion system.
