Abstract
Obesitas merupakan kondisi kronis yang berisiko menimbulkan komplikasi metabolik seperti diabetes melitus tipe 2. Penelitian ini bertujuan untuk mengevaluasi potensi serbuk komponen pembentuk gel cincau hitam (Mesona palustris Blume) dalam menurunkan kadar glukosa darah dan meningkatkan sensitivitas insulin pada mencit obesitas yang diinduksi pakan tinggi lemak. Sebanyak enam kelompok hewan uji digunakan: normal, kontrol negatif (Na-CMC 0,5%), kontrol positif (metformin), serta tiga kelompok perlakuan komponen pembentuk gel dengan dosis 100, 200, dan 300 mg/KgBB. Induksi selama 21 hari berhasil membentuk kondisi obesitas dan hiperglikemia (IMT > 0,3; glukosa > 126 mg/dL). Perlakuan selama 7 hari menunjukkan penurunan kadar glukosa darah, dengan dosis 300 mg/KgBB menghasilkan penurunan sebesar 9,37%, mendekati efektivitas metformin (15,09%). Peningkatan sensitivitas insulin juga diamati melalui nilai konstanta tes toleransi insulin (KTTI), di mana dosis 300 mg/KgBB mencapai 83,36, hampir setara dengan metformin (88,48). Hasil ini menunjukkan bahwa komponen pembentuk gel cincau hitam memiliki potensi sebagai agen antidiabetes alami melalui mekanisme peningkatan sensitivitas insulin dan penurunan kadar glukosa darah pada kondisi obesitas. ----- Obesity is a chronic condition that increases the risk of metabolic complications such as type 2 diabetes mellitus. This study aimed to evaluate the potential of powdered gel-forming components of black grass jelly (Mesona palustris Blume) in lowering blood glucose levels and enhancing insulin sensitivity in obese mice induced by a high-fat diet. Six groups of test animals were used: normal, negative control (0.5% Na-CMC), positive control (metformin), and three treatment groups receiving the gel-forming component at doses of 100, 200, and 300 mg/kg body weight. A 21-day induction period successfully established obesity and hyperglycemia (BMI > 0.3; blood glucose > 126 mg/dL). Seven days of treatment showed a reduction in blood glucose levels, with the 300 mg/kg dose resulting in a 9.37% decrease, approaching the effectiveness of metformin (15.09%). An increase in insulin sensitivity was also observed through the insulin tolerance test constant (ITTC), where the 300 mg/kg dose reached 83.36, nearly equivalent to metformin (88.48). These findings suggest that the gel-forming components of black grass jelly have promising potential as a natural antidiabetic agent through mechanisms of improving insulin sensitivity and lowering blood glucose levels under obese conditions.
