Abstract
Tanaman rumput mutiara (Oldenlandiae corymbosa Lamk.) telah lama digunakan dalam pengobatan tradisional karena kandungan senyawa aktifnya, termasuk yang berpotensi sebagai antioksidan. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi senyawa antioksidan dari fraksi etil asetat ekstrak etanol herba rumput mutiara. Ekstraksi dilakukan melalui metode maserasi dengan pelarut etanol, kemudian dilakukan pemisahan menggunakan fraksinasi cair-cair menggunakan pelarut n-heksan, etil asetat, dan air. Fraksi etil asetat yang menunjukkan aktivitas antioksidan berdasarkan uji DPPH (2,2 -difenil-1- pikrilhidrazil) dipisahkan lebih lanjut menggunakan kromatografi kolom gravitasi dan KLT preparatif untuk memperoleh isolat murni. Uji kemurnian dilakukan dengan KLT dua dimensi, sedangkan identifikasi struktur senyawa dilakukan menggunakan spektrofotometri UV-Vis dengan pereaksi geser serta uji penampak bercak menggunakan AlCl₃ dan uap amonia. Hasil menunjukkan adanya satu noda tunggal dan perubahan warna spesifik yang mengindikasikan keberadaan senyawa golongan fenolik. Senyawa yang berhasil diisolasi dari fraksi etil asetat ekstrak etanol herba rumput mutiara diprediksi termasuk dalam golongan senyawa fenolik dan menunjukkan aktivitas antioksidan yang potensial. ----- Oldenlandiae corymbosa Lamk has long been used in traditional medicine due to its content of bioactive compounds, including those with potential antioxidant properties. This study aimed to isolate and identify antioxidant compounds from the ethyl acetate fraction of the ethanol extract of O. corymbosa herb. Extraction was carried out using the maceration method with ethanol as the solvent, followed by liquid-liquid fractionation using n-hexane, ethyl acetate, and water. The ethyl acetate fraction, which showed antioxidant activity based on the DPPH (2,2 - difenil-1-pikrilhidrazil) assay, was further separated using gravity column chromatography and preparative thin-layer chromatography (TLC) to obtain a pure isolate. Purity testing was conducted using two-dimensional TLC, while structural identification was performed using UV-Vis spectrophotometry with shift reagents, as well as spot detection using AlCl₃ and ammonia vapor. The results revealed a single spot and specific color changes, indicating the presence of phenolic compounds. The compound isolated from the ethyl acetate fraction of the ethanol extract of O. corymbosa is predicted to belong to the phenolic group and demonstrates potential antioxidant activity.
