Abstract
Senyawa bahan alam memiliki potensi untuk dikembangkan sebagai produk veteriner, dalam proses pemanfaatannya diketahui bahwa ampas ekstrak dari isolasi kurkumin (Curcuma longa L.), mangostin (Garcinia mangostana L.), dan piperin (Piper nigrum L.) masih mengandung senyawa bioaktif dengan potensi pemanfaatan ulang. Pada penelitian ini dilakukan analisis senyawa kurkumin, mangostin dan piperin untuk mengetahui kadar senyawa yang terkandung pada masing-masing ampas ekstrak. Preparasi yang dilakukan dengan menggunakan 20 gram ampas ekstrak dalam 80 mL metanol menghasilkan rendemen berturut-turut 0,18% (kurkumin), 0,39% (mangostin), dan 0,065% (piperin). Penetapan panjang gelombang maksimum menunjukkan nilai spesifik: 421 nm (kurkumin), 316 nm (mangostin), dan 331 nm (piperin). Kurva kalibrasi ketiganya menunjukkan linearitas sangat baik (r² > 0,998). Uji akurasi menunjukkan perolehan kembali 89,94–101,19% (kurkumin), 85,80–108,13% (mangostin), dan 99,42–102,58% (piperin), sesuai rentang validasi. Presisi berdasarkan RSD sebagian besar < 5%, dengan nilai tertinggi 8,7773% hanya pada konsentrasi rendah mangostin. Uji spesifisitas menunjukkan kesesuaian spektrum dengan standar senyawa. Nilai LOD dan LOQ menunjukkan sensitivitas metode terhadap konsentrasi rendah. Berdasarkan hasil ini, metode spektrofotometri UV-Visibel terbukti valid untuk analisis ketiga senyawa dalam ampas ekstrak, yang berpotensi dimanfaatkan kembali sebagai sumber bahan aktif dalam pengembangan sediaan farmasi berbasis bahan alam. ----- Natural compounds have the potential to be developed as veterinary products. In the process of utilization, it is known that the extract dregs from the senyawaion of curcumin (Curcuma longa L.), mangostin (Garcinia mangostana L.), and piperine (Piper nigrum L.) still contain bioactive compounds with the potential for reuse. In this study, an analysis of curcumin, mangostin, and piperine compounds was carried out to determine the levels of compounds contained in each extract. Preparation carried out using 20 grams of extract dregs in 80 mL of methanol produced yields of 0.18% (curcumin), 0.39% (mangostin), and 0.065% (piperine), respectively. Determination of the maximum wavelength showed specific values: 421 nm (curcumin), 316 nm (mangostin), and 331 nm (piperine). The calibration curves of the three showed very good linearity (r² > 0.9998). Accuracy tests showed recovery of 89.94–101.19% (curcumin), 85.80–108.13% (mangostin), and 99.42 102.58% (piperine), according to the validation range. Precision based on RSD was mostly <5%, with the highest value of 8,7773% only at low concentrations of mangostin. The specificity test showed the suitability of the spectrum with the senyawae standard. LOD and LOQ values showed the sensitivity of the method to low concentrations. Based on these results, the UV-Visible spectrophotometry method was proven valid for the analysis of the three compounds in the extract pulp, which has the potential to be reused as a source of active ingredients in the development of natural ingredient-based pharmaceutical preparations.
