Abstract

Andrografolid adalah komponen utama dengan berbagai manfaat dalam pengobatan tradisional. Penelitian ini bertujuan untuk mengoptimasi berat silika gel, konsentrasi ekstrak, dan volume ekstrak dalam pemurnian ekstrak sambiloto. Penelitian dilakukan dengan memvariasikan berat silika gel dari 250 mg, 500 mg dan 750 mg. Konsentrasi ekstrak yang digunakan yaitu 300, 500 dan 700 ppm. Sedangkan variasi volume ekstrak yang digunakan yaitu 5 mL, 10 dan 15 mL. Isolat hasil pemurnian dikarakterisasi dengan menggunakan kromatografi lapis tipis, spektrofotometri uv-visibel, dan kromatografi cair kinerja tinggi (KCKT). Berdasarkan hasil penelitian didapatkan bahwa kondisi optimal proses pemurnian didapatkan pada berat adsorben 750 mg, volume ekstrak 5 mL, dan konsentrasi ekstrak 300 ppm. Hasil KLT standar dengan nilai Rf 0,29 dan Sampel Rf 0,12-0,27. Hasil uji kemurnian dengan KCKT menunjukkan peningkatan kemurnian ekstrak setelah dimurnikan pada kondisi optimal dari 74% menjadi 82,60%. --- Andrographolide is a primary component with diverse benefits in traditional medicine. This research aimed to optimize the weight of silica gel, extract concentration, and extract volume in the purification of Andrographis extract. The study was conducted by varying the weight of silica gel at 250 mg, 500 mg, and 750 mg. The extract concentrations used were 300 ppm, 500 ppm, and 700 ppm. Meanwhile, the extract volume variations included 5 mL, 10 mL, and 15 mL. The isolated purification chromatography, products were characterized using thin-layer spectrophotometry, and high-performance liquid chromatography (HPLC). Based on the research findings, optimal purification conditions were achieved with 750 mg adsorbent weight, 5 mL extract volume, and 300 ppm extract concentration. The standard thin-layer chromatography results showed an Rf value of 0.29, while sample Rf ranged from 0.12 to 0.27. The purity test results using HPLC demonstrated an enhancement in the extract purity after purification under optimal conditions, from 74% to 82.60%.
