Abstract
Vaksin Hepatitis B rekombinan dapat diproduksi menggunakan ragi Hansenula polymorpha. Isolasi genom Hansenula polymorpha telah banyak dilakukan dengan metode konvensional. Kit Yeast DNA Purification Masterpure Epicentre merupakan kit yang efektif dan efisien untuk mengisolasi genom ragi namun belum spesifik untuk Hansenula polymorpha. Penelitian ini dilakukan untuk menentukan nilai Optical Density (OD) yang optimum pada isolasi genom ragi Hansenula polymorpha menggunakan kit. Metode diawali dengan variasi kultur Hansenula polymorpha pada OD 6, 8, 10, 12, 14, 16, 18 dan 20. Penentuan kemurnian secara kualitatif dilakukan dengan elektroforesis gel agarosa dan secara kuantitatif menggunakan Nano Drop. Penentuan konsentrasi genom secara kuantitatif dilakukan dengan Qubit fluorometer 2.0. Pada penelitian ini diperoleh nilai konsentrasi genom Hansenula polymorpha tertinggi yaitu 13,399 µg/mL.;---Recombinant Hepatitis B vaccines can be produced using Hansenula polymorpha as a host. Isolation of Hansenula polymorpha genome by conventional methods has been widely used. Yeast DNA Purification Masterpure Epicentre is an effective and efficient kit for isolating yeast genomes but has not yet specific to Hansenula polymorpha. This research was conducted to determine the optimum Optical Density (OD) in Hansenula polymorpha genome that can be isolated using a kit. The method begins with variations in Hansenula polymorpha culture on OD 6, 8, 10, 12, 14, 16, 18 and 20. Purity of the genom qualitatively determined by agarose gel electrophoresis and quantitatively determined using NanoDrop. Concentration of the genom quantitatively determine with fluorometre Qubit 2.0. In this study the highest Hansenula polymorpha genomic concentration was reach 13.399 µg/mL.
