Abstract

Penelitian ini mengfokuskan pada pengembangan sediaan gel orabase berbahan aktif ekstrak kayu secang (Caesalpinia sappan Linn) dan isolat brazilin yang memiliki potensi sebagai antibakteri terhadap Aggregatibacter actinomycetemcomitans dan Porphyromonas gingivalis penyebab penyakit periodontitis. Gel orabase dirancang dengan dua basis yang berbeda yaitu Natrium Karboksimetilselulosa (Na CMC) kombinasi dengan Vaseline Flavum serta Carbopol 940P kombinasi dengan Vaseline Flavum yang dibuat dalam tiga variasi konsentrasi basis. Uji stabilitas fisik sediaan gel dilakuan pada penyimpanan dipercepat pada suhu 40°C ± 2°C RH 75% ± 5% selama 28 hari dengan parameter fisik yang digunakan antara lain organoleptis, homogenitas, pH, sineresis, viskositas, daya sebar, dan daya lekat. Hasil optimasis basis gel formula F2, F3, dan F4 pada basis Na CMC dan formula F3, F4, dan F5 dengan basis Carbopol 940P terpilih sebagai kandidat untuk dibuat sediaan gel orabase. Hasil uji stabilitas fisik menunjukan bahwa gel orabase dengan basis Na CMC memiliki stabilitas paling baik pada pengamatan organoleptis tidak terjadi perubahan fisik, homogenitas tetap homogen baik sebelum dan selama penyimpanan, hasil uji pH tidak terjadi perubahan yang signifikan, tidak terjadi sineresis, serta hasil uji viskositas, daya sebar, dan daya lekat yang stabil. --- This study focuses on the development of an orabase gel preparation made from sappan wood extract (Caesalpinia sappan Linn) and brazilin isolate that has potential as an antibacterial against Aggregatibacter actinomycetemcomitans and Porphyromonas gingivalis that cause periodontitis. The orabase gel was designed with two different bases namely Sodium Carboxymethylcellulose (Na CMC) combined with Vaseline Flavum and Carbopol 940P combined with Vaseline Flavum made in three variations of base concentration. The physical stability test of the gel preparation was carried out in accelerated storage at 40°C ± 2°C RH 75% ± 5% for 28 days with physical parameters used including organoleptic, homogeneity, pH, syneresis, viscosity, spreadability, and stickiness. The gel base optimization results of formulas F2, F3, and F4 on Na CMC base and formulas F3, F4, and F5 on Carbopol 940P base were selected as candidates for making orabase gel preparations. The results of the physical stability test showed that the orabase gel with Na CMC base had the best stability in organoleptic observations, no physical changes occurred, homogeneity remained homogeneous both before and during storage, pH test results did not occur significant changes, no syneresis occurred, and the results of viscosity, spreadability, and stickiness tests were stable.
