Abstract
Karagenan dapat diperoleh dengan proses ekstraksi menggunakan pelarut alkali seperti NaOH, KOH, dan Ca(OH)2. Karagenan yang sudah dimodifikasi secara enzimatis menghasilkan bobot molekul yang lebih kecil sehingga penggunaannya sebagai bahan tambahan obat semakin luas. Tujuan dari penelitian ini yaitu untuk mengaplikasikan karagenan hasil modifikasi enzimatis sebagai bahan pengikat sediaan tablet Klorfeniramin maleat. Metode yang digunakan dalam penelitian ini yaitu uji karakteristik fisika padatan karagenan, formulasi dan pembuatan granul dengan metode granulasi basah, evaluasi masa siap cetak, pencetakan tablet dan evaluasi tablet. Terdapat 9 formula karagenan hasil modifikasi enzimatis (NaOH, KOH, dan Ca(OH)₂) dengan variasi konsentrasi F1 (1,5%); F2 (3%); F3 (6%) serta 1 formula pembanding F4 (5%) (PVP K-30) sebagai bahan pengikat. Hasil evaluasi karakteristik fisika padatan yaitu true density (1,87-1,93), solid fraction (0,20-0,24), tensile strength (0,0556-0,0568), bonding index (0,0108-0,0112), BFI (0,1017- 0,1150) dan seluruh formula memenuhi syarat evaluasi massa siap cetak meliputisusut pengeringan, distribusi ukuran partikel, laju alir, sudut istirahat, kompresibilitas. Pada evaluasi tablet F1 (1,5%) karagenan hasil modifikasi enzimatis NaOH, KOH, dan Ca(OH)₂ tidak memenuhi uji friabilitas dan friksibilitas, F2 (3%); F3 (6%); F4 (5%) memenuhi seluruh evaluasi tablet. Sehingga karagenan hasil modifikasi enzimatis NaOH, KOH, dan Ca(OH)₂ dapat digunakan sebagai bahan pengikat. --- Carrageenan can be obtained by extraction process using alkaline solvents such as NaOH, KOH, and Ca(OH)2. Enzymatically modified carrageenan produces a smaller molecular weight so that its use as a drug additive is increasingly widespread. The purpose of this study was to apply enzymatically modified carrageenan as a binder for Klorfeniramin maleate tablet preparation. The methods used in this study were testing the physical characteristics of carrageenan solids, formulating and making granules using the wet granulation method, evaluating the press-ready period, tablet molding and tablet evaluation. There are 9 formulas of enzymatically modified carrageenan (NaOH, KOH, and Ca(OH)₂) with varying concentrations of F1 (1.5%); F2 (3%); F3 (6%) and 1 comparison formula F4 (5%) (PVP K-30) as a binder. The results of the evaluation of the physical characteristics of solids are true density (1.87-1.93), solid fraction (0.20- 0.24), tensile strength (0.0556-0.0568), bonding index (0.0108-0.0112), BFI (0.1017-0.1150) and all formulas meet the evaluation requirements of ready-to press masses, namely drying shrinkage, particle size distribution, flow rate, angle of repose, compressibility. In the evaluation of tablets F1 (1.5%), the enzymatically modified carrageenan NaOH, KOH, and Ca(OH)₂ did not meet the friability and fricibility tests, F2 (3%); F3 (6%); F4 (5%) met all tablet evaluations. So that the enzymatically modified carrageenan NaOH, KOH, and Ca(OH)₂ can be used as an ingredient in tablets.
