Abstract
Biosurfaktan adalah molekul aktif permukaan yang berasal dari sel-sel hidup. Biosurfaktan yang dihasilkan oleh mikroorganisme merupakan zat yang mampu menurunkan tegangan permukaan, dan berasal dari berbagai jenis mikroorganisme. Sumber mikroorganisme ini dapat berasal dari berbagai lingkungan, salah satunya bakteri pada VCO. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri penghasil biosurfaktan dari Virgin Coconut Oil (VCO). VCO dibuat melalui fermentasi alami santan kelapa. Isolasi bakteri dilakukan dengan metode spread plate pada media NA. Karakterisasi isolat meliputi pewarnaan gram, uji fermentasi, pertumbuhan pada variasi suhu dan konsentrasi NaCl, serta uji penapisan pada media blood agar. Aktivitas biosurfaktan diuji dengan metode emulsifikasi, oil spreading, dan drop collapse. Hasil menunjukkan isolat bakteri dari VCO bersifat gram positif dan homofermentatif, dengan pertumbuhan optimal pada 37°C dan toleransi terhadap 6,5% NaCl. Uji penapisan mengonfirmasi sifat non-patogen isolat. Aktivitas biosurfaktan terdeteksi terutama pada konsentrasi isolat 10−4 (3)1. Dari hasil penelitian dapat disimpulkan bahwa VCO mengandung bakteri yang dapat menghasilkan biosurfaktan, dilihat dari karakteristik mikroskopik berbentuk batang dan berwarna ungu. ----- Biosurfactants are surface-active molecules derived from living cells. Biosurfactants produced by microorganisms are substances capable of reducing surface tension and originate from various types of microorganisms. These microorganisms can be sourced from diverse environments, one of which is bacteria found in Virgin Coconut Oil (VCO). This study aims to isolate and identify biosurfactant-producing bacteria from VCO. VCO was produced through the natural fermentation of coconut milk. Bacterial isolation was performed using the spread plate method on NA media. The characterization of the isolates included Gram staining, fermentation tests, growth at varying temperatures and NaCl concentrations, as well as screening tests on blood agar media. Biosurfactant activity was tested using emulsification, oil spreading, and drop collapse methods. The results showed that the bacterial isolates from VCO were Gram-positive and homofermentative, with optimal growth at 37°C and tolerance to 6.5% NaCl. Screening tests confirmed the non-pathogenic nature of the isolates. Biosurfactant activity was primarily detected at an isolate concentration of 10-4 (3)1. Based on the research findings, it can be concluded that VCO contains bacteria capable of producing biosurfactants, as evidenced by their microscopic rod shape and purple color.
