Abstract
Mahkota dewa (Phaleria macrocarpa (Scheff.) Boerl) adalah tanaman yang diketahui mengandung berbagai senyawa bioaktif, salah satunya yaitu flavonoid yang memiliki potensi sebagai antioksidan. Flavonoid berfungsi untuk melindungi tubuh dari kerusakan yang disebabkan oleh radikal bebas yang dapat berkontribusi terhadap berbagai penyakit degeneratif. Penelitian ini bertujuan untuk menentukan kadar total flavonoid dalam ekstrak etanol buah mahkota dewa serta menguji aktivitas antioksidannya menggunakan spektrofotometer UV-Vis dengan metode DPPH (2,2-diphenyl-1-picrylhydrazl) sebagai radikal bebas, metode DPPH memiliki kelebihan yaitu mudah, sederhana, dan lebih efektif dibandingkan metode lain. Proses ekstraksi dilakukan dengan menggunakan metode maserasi dengan pelarut etanol 96%. Hasil penelitian menunjukkan bahwa kadar flavonoid total dalam ekstrak etanol buah mahkota dewa adalah 2,312 mgQE/g ekstrak. Sedangkan uji aktivitas antioksidan dengan metode DPPH menghasilkan nilai IC50 sebesar 340,26 ppm, yang menunjukkan bahwa ekstrak etanol buah mahkota dewa termasuk dalam kategori antioksidan sangat lemah. ------ Mahkota dewa (Phaleria macrocarpa (Scheff.) Boerl) was known to contain various bioactive compounds, one of which was flavonoids, which had potential as antioxidants. Flavonoids functioned to protect the body from damage caused by free radicals that could contribute to various degenerative diseases. This study aimed to determine the total flavonoid content in the ethanol extract of mahkota dewa fruit and test its antioxidant activity using a UV-Vis spectrophotometer with the DPPH (2,2-diphenyl-1-picrylhydrazyl) method as free radicals. The DPPH method has advantages such as being easy, simple, and more effective compared to other methods. The extraction process was conducted using the maceration method with 96% ethanol as the solvent. The results of the study showed that the total flavonoid content in the ethanol extract of mahkota dewa fruit was 2.312 mgQE/g extract. Meanwhile, the antioxidant activity test using the DPPH method produced an IC50 value of 340.26 ppm, indicating that the ethanol extract of mahkota dewa was categorized as a very weak antioxidant.
