Abstract
Potensi katekin dari gambir sebagai antioksidan kuat yang dapat digunakan dalam formulasi sheet mask, sebuah sediaan topikal yang efektif meningkatkan penyerapan zat aktif pada kulit. Penelitian ini bertujuan untuk menguji aktivitas antioksidan, stabilitas dan cemaran mikroba pada essence yang digunakan dalam sediaan sheet mask isolat katekin dari tanaman gambir (Uncaria gambir Roxb.). Pengujian aktivitas antioksidan isolat katekin dilakukan menggunakan metode DPPH, formulasi essence sediaan sheet mask, uji stabilitas sediaan essence sheet mask, pengujian aktivitas antioksidan sediaan essence, uji cemaran mikroba (ALT dan AKK), dan pengujian kadar sediaan essence dibuat 2 formulasi yaitu F1 dan F2 dengan formula basis 2:1. Hasil penelitian menunjukkan bahwa nilai IC50 asam askorbat 3,879 ppm, sedangkan untuk nilai IC50 isolat katekin 3,010 ppm. Formula essence yang paling stabil adalah F1 dengan gelling agent xanthan gum pada konsentrasi 0,2%. Aktivitas antioksidan essence sheet mask pada F1 memiliki nilai IC50 sebesar 64,066 ppm pada hari ke-0 dan 96,049 ppm pada hari ke-28 yang merupakan nilai antioksidan yang kuat. Uji cemaran mikroba menunjukan bahwa hasil ALT dan AKK pada F1 dan F2 berada dibawah batas maksimum (<103 ), serta hasil uji Staphylococcus aureus dan Pseudomonas aeuruginosa menunjukan hasil negatif. Hasil dari keseluruhan nya, essence isolat katekin memenuhi standar stabilitas fisika dan cemaran mikroba. ----- The potential of catechins from gambir as a strong antioxidant that can be used in sheet mask formulations, a topical preparation that effectively increases the absorption of active substances in the skin. This study aims to test the antioxidant activity, stability and microbial contamination of the essence used in sheet mask preparations of catechin isolate from gambier (Uncaria gambir Roxb.). Testing the antioxidant activity of catechin isolates was carried out using the DPPH method, formulation of essence sheet mask preparations, stability testing of essence sheet mask preparations, testing the antioxidant activity of essence preparations, testing microbial contamination (TYMC and TPC), and testing the level of essence preparations made 2 formulations, namely F1 and F2 with a 2:1 base formula. The results showed that the IC50 value of ascorbic acid was 3.879 ppm, while the IC50 value of catechin isolate was 3.010 ppm. The most stable essence formula was F1 with gelling agent xanthan gum at 0.2% concentration. The antioxidant activity of the essence sheet mask in F1 has an IC50 value of 64.066 ppm on day 0 and 96.049 ppm on day 28 which is a strong antioxidant value. The microbial contamination test showed that the TPC and TYMC results in F1 and F2 were below the maximum limit (<103 ), and the Staphylococcus aureus and Pseudomonas aeuruginosa test results showed negative results. Overall, the catechin isolate essence met the physical stability and microbial contamination standards.
