Abstract

Andrografolid merupakan senyawa utama yang terdapat pada tanaman sambiloto (Andrographis paniculata Nees) dan memiliki berbagai aktivitas farmakologis, seperti anti-inflamasi, anti-aterosklerotik, antioksidan, anti-hiperglikemik, dan antimalaria. Penelitian ini bertujuan untuk mengevaluasi keamanan dan potensi efek toksik dari isolat andrografolid melalui kajian histopatologis pada mencit Swiss Webster. Uji dilakukan dengan pemberian oral selama 28 hari menggunakan metode OECD 407. Mencit dibagi menjadi enam kelompok uji, yaitu kontrol normal, dosis rendah (5 mg/kgBB), dosis menengah (500 mg/kgBB), dosis tinggi (1000 mg/kgBB), kelompok satelit (1000 mg/kgBB), dan kontrol satelit. Hasil pengamatan makroskopik menunjukkan bahwa pemberian isolat andrografolid tidak menimbulkan gejala toksik yang nyata. Namun, pengamatan mikroskopik mengungkapkan adanya indikasi kelainan ringan pada beberapa organ hati, jantung, limpa, ginjal, paru-paru dan lambung. Meskipun perubahan histopatologis ini masih terkategorikan ringan, penelitian lebih lanjut dengan perpanjangan waktu pemberian menjadi prioritas untuk memastikan keamanan jangka panjang isolat andrografolid. Temuan ini memberikan informasi awal yang berharga mengenai profil toksisitas andrografolid, namun studi lanjutan diperlukan untuk memperkuat data keamanan pada dosis tinggi dan penggunaan jangka panjang. ----- Andrographolide is the main compound found in the Andrographis paniculata Nees plant and has various pharmacological activities, including anti-inflammatory, anti-atherosclerotic, antioxidant, anti-hyperglycemic, and anti-malarial properties. This study aims to evaluate the safety and potential toxic effects of andrographolide isolate through histopathological examination in Swiss Webster mice. The test was conducted with oral administration over 28 days using the OECD 407 method. Mice were divided into six test groups: normal control, low dose (5 mg/kg BW), medium dose (500 mg/kg BW), high dose (1000 mg/kg BW), satellite group (1000 mg/kg BW), and satellite control. Macroscopic observations indicated that the administration of andrographolide isolate did not produce any significant toxic symptoms. However, microscopic examination revealed mild abnormalities in several organs, including the liver, heart, spleen, kidneys, lungs, and stomach. Although these histopathological changes were categorized as mild, further research with an extended treatment duration is prioritized to ensure the long-term safety of andrographolide isolate. These findings provide valuable preliminary information regarding the toxicity profile of andrographolide, but additional studies are needed to strengthen the safety data at higher doses and prolonged use.
