Abstract

Flavonoid merupakan suatu senyawa metabolit sekunder yang terbesar pada tumbuhan yang memiliki potensi sebagai antioksidan, tanaman yang diketahui memiliki kandungan senyawa flavonoid dan potensi antioksidan adalah daun kirinyuh. Tujuan dari penelitian ini untuk mengetahui kadar total flavonoid dan aktivitas antioksidan pada daun kirinyuh. Kadar flavonoid total yang diperoleh berturut-turut dari replikasi 1 sebesar 2,18 mgQE/g ekstrak, replikasi 2 sebesar 3,86 mgQE/g ekstrak, dan replikasi 3 sebesar 6,34 mgQE/g ekstrak. Untuk penentuan aktivitas antioksidan pembanding yang digunakan pada penelitian ini adalah vitamin C. Pada pengujian ini larutan vitamin C dibuat dengan beberapa konsentrasi yaitu 2, 4, 6, 8, 10 ppm untuk mendapatkan persamaan regresi linier. Didapatkan persamaan garis y = 7,0001x – 5,3351 dan diperoleh nilai IC sebesar 6,38 ppm sehingga dapat dikatakan aktivitas antioksidannya sangat kuat. Dari masing-masing konsentrasi sampel uji menggunakan spektrofotometer UV-Vis didapatkan persentase inhibisi (%) hasil tersebut diplotkan dalam sebuah grafik dan didapatkan persamaan y = 0,216x – 39,478 dan nilai IC50 yang diperoleh sebesar 48,71 ppm. Berdasarkan hasil penelitian yang dilakukan dapat disimpulkan ekstrak dengan kadar flavonoid tertinggi terdapat pada replikasi ke 3 sebesar 6,34 mgQE/g ekstrak. Dan hasil dari penentuan aktivitas antioksidan ekstrak daun kirinyuh menggunakan metode DPPH menunjukkan bahwa nilai IC50 yang diperoleh sebsar 48,71 ppm sehingga dapat dinyatakan memiliki aktivitas antioksidan yang sangat kuat. ---- Flavonoids are the largest secondary metabolite compounds in plants which have potential as antioxidants. The plant which is known to contain flavonoid compounds and antioxidant potential is kirinyuh leaves. The aim of this research was to determine the total levels of flavonoids and antioxidant activity in kirinyuh leaves. Total flavonoid levels obtained respectively from replication 1 were 2.18 mgQE/g extract, replication 2 was 3.86 mgQE/g extract, and replication 3 was 6.34 mgQE/g extract. To determine antioxidant activity, the comparison used in this research was vitamin C. In this test, vitamin C solutions were made with several concentrations, namely 2, 4, 6, 8, 10 ppm to obtain a linear regression equation. The line equation y = 7.0001x – 5.3351 was obtained and an IC value of 6.38 ppm was obtained, so it can be said that the antioxidant activity is very strong. From each concentration of the test sample using a UV-Vis spectrophotometer, the percentage of inhibition (%) was plotted in a graph and the equation obtained was y = 0.216x – 39.478 and the IC50 value obtained was 48.71 ppm. Based on the results of the research conducted, it can be concluded that the extract with the highest flavonoid content was found in the 3rd replication at 6.34 mgQE/g extract. And the results of determining the antioxidant activity of kirinyuh leaf extract using the DPPH method show that the IC50 value obtained is 48.71 ppm so that it can be stated to have very strong antioxidant activity
