Abstract

Andrografolid merupakan senyawa aktif yang berasal dari tanaman sambiloto [Andrographis paniculata (Burm.F.)] telah dikenal memiliki berbagai aktivitas farmakologis, termasuk potensi sebagai agen anti-kanker. Penelitian ini bertujuan untuk menganalisis jalur kematian sel kanker paru A-549 yang diinduksi oleh isolat andrografolid menggunakan metode In-Cell Western. Dalam penelitian ini, andrografolid diuji pada sel kanker paru A-549 untuk mengukur aktivitas apoptosis melalui peningkatan ekspresi Caspase-3, sebuah enzim kunci dalam jalur kematian sel terprogram. Variasi dosis yang digunakan adalah sebanyak 4 dosis, dimulai dari dosis ½ x IC50, 1x IC50, 2x IC50 dan 3 x IC50. Hasil penelitian menunjukkan bahwa pemberian andrografolid mampu meningkatkan ekspresi Caspase-3, terutama pada dosis 2x IC50 sebanyak 95,56 µg/mL dan 3x IC50 sebanyak 141,84 µg/mL yang menunjukkan aktivitas jalur apoptosis yang signifikan. Selain itu, perubahan morfologi sel yang diamati mengkonfirmasi adanya kematian sel pada konsentrasi andrografolid yang lebih tinggi, serupa dengan efek agen kemoterapi doxorubicinHCl. Dengan demikian, andrografolid menunjukkan potensi besar sebagai agen anti-kanker melalui induksi apoptosis pada sel kanker paru A-549. ----- Andrographolide, an active compound derived from the Andrographis paniculata (Burm.F.) plant, is known for its various pharmacological activities, including its potential as an anti-cancer agent. This study aims to analyze the cell death pathway of A-549 lung cancer cells induced by andrographolide isolate using the In-Cell Western method. In this research, andrographolide was tested on A-549 lung cancer cells to measure apoptosis activity through the increased expression of Caspase-3, a key enzyme in the programmed cell death pathway. Four dosage variations were used, starting from ½ x IC50, 1x IC50, 2x IC50, and 3x IC50. The results showed that the administration of andrographolide significantly increased Caspase-3 expression, particularly at doses of 2x IC50 (95.56 µg/mL) and 3x IC50 (141.84 µg/mL), indicating significant activation of the apoptosis pathway. Additionally, the observed changes in cell morphology confirmed cell death at higher andrographolide concentrations, similar to the effects of the chemotherapeutic agent doxorubicin HCl. Thus, andrographolide shows great potential as an anti-cancer agent by inducing apoptosis in A-549 lung cancer cells.
