Abstract

Isolat andrografolid adalah senyawa bioaktif yang diperoleh dari tanaman Andrographis paniculata, yang dikenal memiliki berbagai manfaat kesehatan. Penelitian ini bertujuan untuk mengevaluasi tingkat keamanan dan efek toksisitas subkronis dari pemberian isolat andrografolid terhadap profil hematologi mencit Swiss Webster selama 28 hari, serta untuk menentukan dosis isolat andrografolid yang dapat menimbulkan efek hematologis dengan menggunakan metode OECD-407. Dalam penelitian ini, mencit dibagi menjadi enam kelompok uji, yaitu kelompok kontrol, dosis 5 mg/kg BB, dosis 500 mg/kg BB, dosis 1000 mg/kg BB, kelompok kontrol satelit, dan kelompok satelit dosis 1000 mg/kg BB. Hasil pengukuran parameter hematologi, termasuk eritrosit, leukosit, hemoglobin, hematokrit, MCV, MCH, MCHC, platelet, neutrofil, dan limfosit, isolat andrografolid tidak menunjukkan kelainan selama 28 hari pemberian. Meskipun terdapat variasi dalam bobot badan mencit, isolat andrografolid dengan dosis 5 mg/kg BB, 500 mg/kg BB, dan 1000 mg/kg BB tidak menimbulkan efek toksik terhadap mencit jantan dan betina sebagai hewan uji. Hasil penelitian menunjukkan bahwa pemberian isolat andrografolid oral selama 28 hari tidak memberikan dampak negatif terhadap profil hematologi. ----- Andrographolide isolate is a bioactive compound derived from the Andrographis paniculata plant, known for its various health benefits. This study aims to evaluate the safety and subchronic toxicity effects of andrographolide isolate on the hematological profile of Swiss Webster mice after a 28-day treatment, as well as to determine the dose of andrographolide isolate that may induce hematological effects by using OECD-407 method. In this study, the mice were divided into six test groups: a control group, the test group dose of 5 mg/kgBW, 500 mg/kgBW, 1000 mg/kgBW, satellite control, and satellite 1000 mg/kg BW. Measurements of hematological parameters, including erythrocytes, leukocytes, hemoglobin, hematocrit, MCV, MCH, MCHC, platelets, neutrophils, and lymphocytes, indicated that andrographolide isolate showed no abnormalities over the 28-days. Even though there were variations in the body weight of mice, andrographolide isolates at doses of 5 mg/kgBW, 500 mg/kgBW and 1000 mg/kgBW did not cause toxic effects on male and female mice as test animals. The results showed that the administration of oral andrographolide isolates for 28 days did not harm the hematological profile.
