Abstract

Daun Binahong (Anredera cordifolia (Tenore) Steenis) merupakan salah satu tanaman yang memiliki aktivitas gastroprotektif dengan kandungan senyawa berkhasiat flavonoid dan tanin. Kurkumin merupakan senyawa polifenol yang dapat ditemukan pada ekstrak temulawak, temugiring, dan kunyit yang juga mempunyai aktivitas gastroprotektif, dikarenakan sifatnya yang menurunkan sekresi asam lambung dan meningkatkan mekanisme pertahanan mukosa. Penelitian ini bertujuan untuk mengetahui dosis dari kombinasi ekstrak daun binahong dan kurkumin yang efektif sebagai gastroprotektif. Simplisia daun binahong diekstraksi dengan menggunakan metode dekoktasi dan dikeringkan dengan menggunakan freeze dryer. Pengujian tukak lambung dilakukan pada tikus wistar jantan. Tikus dipuasakan selama 18 jam dan dibagi menjadi 9 kelompok dengan jumlah setiap kelompok 3 ekor tikus yang terdiri dari kontrol normal, kontrol negatif, kontrol positif (sukralfat 45 mg/kgBB), ekstrak daun binahong (EDB) 200 mg/kgBB, kurkumin (K) 5 mg/kgBB, EDB:K 100:2,5 mg/kgBB, EDB:K 100:5 mg/kgBB, EDB:K 200:2,5 mg/kgBB, dan EDB:K 200:5 mg/kgBB po. Semua kelompok tikus kecuali kelompok normal diinduksi dengan etanol 80% 5 mL/kgBB. Setelah 1 jam tikus diberikan perlakuan sesuai kelompoknya. Setelah 4 jam tikus di euthanasia lalu di bedah dan diambil lambungnya untuk dilakukan pengamatan secara makroskopik dan mikroskopik. Data meliputi pH lambung, jumlah tukak, skor keparahan tukak, persen inhibisi tukak serta skoring histologi lambung. Data dianalisis secara statistik menggunakan ANOVA (Analysis of Variance) dengan signifikansi (p<0,05). Hasil penelitian menunjukkan bahwa kombinasi ekstrak daun binahong 200 mg/kgBB dan kurkumin 5 mg/kgBB memiliki aktifitas gastroprotektif yang paling baik dibandingkan dengan kelompok dosis tunggalnya, dengan persen inhibisi tukak 28,77% dan hasil profil histopatologi menunjukkan bahwa dengan adanya dosis kombinasi tidak terjadi ulserasi epitel. --- The leaves of Binahong (Anredera cordifolia (Tenore) Steenis) are a plant that has gastroprotective activity and contains flavonoids and tannins. Curcumin is a polyphenol compound that can be found in the extracts of ginger, temugiring, and turmeric which also has gastroprotective activity, due to its properties that reduce gastric acid secretion and increase mucosal defense mechanisms. This study aims to determine the dosage of the combination of binahong leaf extract and curcumin which is effective as a gastroprotective. Binahong leaf simplicia was extracted using the decoction method and dried using a freeze dryer. Gastric ulcer testing was carried out on male Wistar rats. The rats were fasted for 18 hours and divided into 9 groups with 3 mice each consisting of normal control, negative control, positive control (sucralfate 45 mg/kgBW), binahong leaf extract (BLE) 200 mg/kgBW, curcumin (C) 5 mg/kgBW, BLE:C 100:2.5 mg/kg, BLE:C 100:5 mg/kg, BLE:C 200:2.5 mg/kg, and BLE:C 200:5 mg/kgBW po. All groups of mice except the normal group were induced with 80% ethanol 5 mL/kgBW. After 1 hour the rats were given treatment according to the group. After 4 hours of euthanasia, the rats were surgically removed and their stomachs were taken for macroscopic and microscopic observation. Data included gastric pH, number of ulcers, ulcer severity score, percent ulcer inhibition, and gastric histological scoring. Data were analyzed statistically using ANOVA (Analysis of Variance) with significance (p <0.05). The results showed that the combination of 200 mg/kgBW binahong leaf extract and 5 mg/kgBW curcumin had the best gastroprotective activity compared to the single-dose group, with percent ulcers inhibition 28.77% and the histopathological profile results showed that the combination dose did not epithelial ulceration occurs.
