Abstract
Bakteri endofit merupakan bakteri yang hidup dalam jaringan tanaman inang tanpa menyebabkan gejala penyakit dan dapat menghasilkan metabolit sekunder sama dengan tanaman inangnya, hal tersebut menjadi peluang yang sangat besar untuk memproduksi metabolit sekunder dari bakteri. Salah satu tanaman yang sudah banyak dimanfaatkan secara tradisional dan telah banyak dibuktikan memiliki berbagai aktivitas biologis adalah daun kumis kucing. Penelitian ini bertujuan untuk mengisolasi bakteri endofit dari daun kumis kucing dan mengidentifikasi metabolit sekundernya. Bakteri endofit diisolasi dari daun kumis kucing dengan metode sebar pada media Trypticase Soy Agar (TSA) pada suhu ruang. Isolat bakteri endofit kemudian diidentifikasi menggunakan sekuensing gen 16S rRNA. Metabolit sekunder yang diproduksi oleh bakteri endofit diidentifikasi dengan cara penapisan fitokimia. Hasil isolasi menunjukkan terdapat 3 bakteri endofit pada daun kumis kucing yaitu Acinetobacter schindleri, Pantoea agglomerans, dan Pseudomonas lurida. Bakteri endofit Pantoea agglomerans menghasilkan senyawa fenolat, sedangkan dua bakteri lainnya tidak teridentifikasi menghasilkan senyawa metabolit sekunder. --- Endophytic bacteria are bacteria that live in the tissue of the host plant without causing symptoms of disease and can produce secondary metabolites similar to that of the host plant, this is a very big opportunity to produce secondary metabolites from bacteria. One plant that has been widely used traditionally and has been proven to have various biological activities is cat whiskers leaves. This study aims to isolate endophytic bacteria from cat whiskers and identify their secondary metabolites. Endophytic bacteria were isolated from cat whiskers using spread plate method on Trypticase Soy Agar (TSA) medium at room temperature. Endophytic bacterial isolates were then identified using 16S rRNA gene sequencing. The secondary metabolites produced by endophytic bacteria were identified by phytochemical screening. The results of the isolation showed that there were 3 endophytic bacteria on cat whiskers, namely Acinetobacter schindleri, Pantoea agglomerans, and Pseudomonas lurida. Pantoea agglomerans endophytic bacteria produce phenolic compounds, while the other two bacteria do not identified produce secondary metabolites.
