Abstract
Kabau (Archidendron bubalinum (Jack) I.C. Nielsen) secara empiris diketahui memiliki manfaat untuk kesehatan diantaranya memiliki aktivitas antioksidan dan mengobati diabetes. Biji merupakan salah satu komponen yang terdapat pada tumbuhan kabau (Archidendron bubalinum (Jack) I.C. Nielsen) yang memiliki kandungan fenol dan flavonoid didalamnya. Penelitian ini bertujuan untuk menentukan kadar total flavonoid dan fenol pada ekstrak biji kabau dari metode ekstraksi refluks dan infundasi. Ekstraksi dilakukan dengan menggunakan metode refluks dengan pelarut etanol 96% dan infundasi dengan pelarut aquadest. Hasil penapisan fitokimia ekstrak biji kabau mengandung senyawa flavonoid, fenolat, tanin, kuinon, saponin, alkaloid, monoterpen, triterpenoid, dan steroid. Pengujian kadar total flavonoid menggunakan reagen aluminium klorida dan kadar total fenol menggunakan reagen Folin–Ciocalteu. Penetapan kadar total diukur dengan menggunakan spektrofotometer UV-Vis. Hasil penelitian menunjukkan bahwa adanya perbedaan metode ekstraksi pada biji kabau dari daerah Sumatera Selatan menyebabkan perbedaan kadar total flavonoid maupun kadar total fenol. Pada metode refluks dan infundasi diperoleh kadar total flavonoid secara berturut-turut yaitu 5,75 ± 0,033 g QE / 100 g dan 1,778 ± 0,030 g QE / 100 g sedangkan pada metode refluks dan infundasi diperoleh kadar total fenol secara berturut-turut yaitu 6,143 ± 0,121 g GAE / 100 g dan 4,128 ± 0,028 g GAE / 100 g. --- Kabau (Archidendron bubalinum (Jack) I.C. Nielsen) is empirically known to have health benefits including antioxidant activity and treating diabetes. Seeds are one of the components found in kabau plants (Archidendron bubalinum (Jack) I.C. Nielsen) which contain phenols and flavonoids in them. This study aims to determine the total levels of flavonoids and phenols in kabau seed extract from reflux and infundation extraction methods. Extraction was carried out by reflux method with ethanol 96% solvent and infundation with aquadest solvent. The results of phytochemical screening for kabau seed extract contained compounds flavonoids, phenolics, tannins, quinones, saponins, alkaloids, monoterpenes, triterpenoids, and steroids. Testing of total flavonoid levels used aluminium chloride reagent and total phenol content using Folin-Ciocalteu reagent. Determination of total content was measured using a spectrophotometer UV-Vis. The results showed that the differences in the extraction method of kabau seeds from the South Sumatera region caused differences in the total flavonoid levels and the total phenol content. In the reflux and infundation methods, the total levels of flavonoids were obtained respectively, namely 5,75 ± 0,033 g QE / 100 g and 1,778 ± 0,030 g QE / 100 g, while in the reflux and infundation methods the total phenol levels were respectively 6,143 ± 0,121 g GAE / 100 g and 4,128 ± 0,028 g GAE / 100 g.
