Abstract
Biosurfaktan adalah produk metabolit yang diproduksi oleh mikroorganisme yang memiliki aktivitas sebagai penurun tegangan permukaan. Bacillus cereus dan Brevundimonas terrae adalah bakteri yang berpotensi digunakan sebagai penghasil biosurfaktan. Upaya optimalisasi biaya bahan baku produksi biosurfaktan adalah dengan pemilihan substrat dengan harga rendah sebagai sumber karbon yaitu minyak jelantah. Penelitian ini bertujuan untuk mendapatkan kondisi optimum produksi biosurfaktan dari Bacillus cereus dan Brevundimonas terrae dengan sumber karbon minyak jelantah. Optimasi produksi biosurfaktan dilakukan pada variasi konsentrasi minyak jelantah yaitu 2%, 3%, 4%, 5% dan variasi pH media 6, 7, 8 serta dilakukan uji aktivitas menggunakan metode indeks emulsifikasi 24 jam (IE24). Penelitian ini menggunakan media produksi Mineral Salt Medium (MSM), konsentrasi inokulum sebanyak 10%, inkubasi pada suhu ruang dengan kecepatan agitasi 160 rpm. Ekstraksi biosurfaktan menggunakan pelarut kloroform : metanol (2:1). Konsentrasi minyak jelantah terbaik pada bakteri Bacillus cereus dan Brevundimonas terrae adalah 3% dan pH terbaik masing- masing adalah pH 6 dan 7. Hasil produksi biosurfaktan dari bakteri Bacillus cereus adalah 8,9 g/L dengan nilai IE24 57,17% dan bakteri Brevundimonas terrae sebanyak 9,63 g/L dengan nilai IE24 61,25%. --- Biosurfatant is metabolite product surface active compound produced by microorganism. Bacillus cereus and Brevundimonas terrae is potential microorganism to produce biosurfactant. One of the effort to minimize the production cost of biosurfactant was selecting low-priced carbon source as a substrate, such as waste frying oil. The aim of this study was to obtain the optimum conditions of biosurfactant produced from Bacillus cereus and Brevundimonas terrae on waste frying oil as the carbon source. The optimization of biosurfactant production was obtained with waste frying concentration as much as 2%, 3%, 4%, and 5%; whereas the medium ph variations were as much as 6, 7, 8; and carried out the biosurfactant activity test used the emulsification index (IE24). Mineral Salt Medium (MSM) was use as production medium, inoculum consentration was 10% and incubation in room temparature with agitation 160 rpm. The biosurfactant extraction was using klorofom : metanol (2:1) as the solvent. The most optimum waste frying oil concentration to produce biosurfactant on Bacillus cereus and Brevundimonas terrae was at 3% and the best optimum medium pH was at pH 6 and 7. Production of crude biosurfactant from Bacillus cereus was as much as 8,9 g/L with IE24 57,17% and Brevundimonas terrae was 9,63 g/L with IE24 61,25%.
