Abstract
Daun kumis kucing (Orthosiphon stamineus Benth.) dan daun seledri (Apium graveolens L.) merupakan tanaman yang mempunyai khasiat sebagai diuretik. Penelitian ini bertujuan untuk mengetahui dosis efektif kombinasi ekstrak daun kumis kucing (Orthosiphon stamineus Benth.) dan daun seledri (Apium graveolens L.) pada tikus putih jantan galur wistar. Hewan uji yang digunakan sebanyak 10 ekor uji pendahuluan dan 25 ekor untuk pengujian aktifitas diuretik. Uji aktifitas diuretik dibagi menjadi 5 kelompok perlakuan yaitu kelompok kontrol negatif (suspensi CMC 0,5%), kelompok kontrol positif (Furosemide 40 mg/kgBB), kelompok uji 1 kombinasi 62,5 mg/kgBB ekstrak daun kumis kucing dan 140 mg/kgBB ekstrak daun seledri, kelompok uji 2 kombinasi 62,5 mg/kgBB ekstrak daun kumis kucing dan 280 mg/kgBB ekstrak daun seledri, kelompok uji 3 kombinasi 62,5 mg/kgBB ekstrak daun kumis kucing dan 420 mg/kgBB ekstrak daun seledri. Volume urin hewan uji diukur setelah perlakuan pada jam ke 1, 2, 3, 4, 5 dan 6 jam. Kemudian dilakukan pengamatan parameter urin meliputi pH, warna kejernihan, dan bobot jenis. Pengolahan data dilakukan dengan menghitung data persen diuretik serta uji statistik ANOVA. Hasil penelitian menunjukan bahwa kelompok uji 2 dengan dosis kombinasi 62,5 mg/kgBB ekstrak daun kumis kucing dan 280 mg/kgBB ekstrak daun seledri memiliki aktivitas diuretik yang mendekati dengan kontrol positif. --- The leaves of cat's whiskers (Orthosiphon stamineus Benth.) and celery leaves (Apium graveolens L,) are plants that have diuretic activity. This study aims to determine the effective dose of a combination of cat whiskers (Orthosiphon stamineus Benth.) and celery (Apium graveolens L.) leaf extract on male white rats Wistar strain. The test animals used for preliminary test are 10 rats and 25 rats for testing diuretic activity. The diuretic activity test was divided into 5 treatment groups, namely a negative control group (CMC suspension 0.5%), a positive control group (Furosemide 40 mg/kgBW), a combination test group 1 combination 62.5 mg / kgBW cat whiskers leaf extract and 140 mg / kgBW of celery leaf extract, test group 2 combination of 62.5 mg / kgBW cat's whiskers leaf extract and 280 mg / kgBW of celery leaf extract, test group 3 combination 62.5 mg / kgBW cat's whiskers leaf extract and 420 mg / kgBW of celery leaf extract . The urine volume of test animals was measured after treatment at 1, 2, 3, 4, 5 and 6 hours. Then the urine parameters were observed including pH, clarity, and specific gravity. Processing data was done by calculating the percent diuretic data and the ANOVA statistical test. The results showed that the second test group with a combined dose of 62.5 mg / kgBW cat's whiskers leaf extract and 280 mg / kgBW celery leaf extract had diuretic activity approaching to the positive control.
