Abstract
Biji Kabau (Achidendron bubalinum (Jack) I.C Nielsen) merupakan salah satu tanaman yang berkhasiat sebagai antidiabetes. Secara empiris di Sumatra Utara tanaman kabau telah digunakan sebagai obat tradisonal antidiabetes herbal, selain itu biji kabau dimanfaatkan sebagai obat untuk sakit perut dan demam. Penelitian ini dilakukan untuk mengetahui aktivitas antidiabetes dari ekstrak etanol 96% dan fraksi metanol 100% biji kabau dengan metode beban glukosa. Penelitian ini menggunakan mencit putih jantan galur Swiss Webster yang dibagi ke dalam 5 kelompok perlakuan, yaitu kontrol normal, kontrol positif (glibenklamid 0,013mg/20g BB mencit), kontrol negatif (Na-CMC 0,5%), ekstrak etanol 96% (250mg/Kg BB mencit) dan fraksi metanol 100% (250mg/Kg BB mencit). Hasil penelitian menunjukkan penurunan kadar glukosa darah relatif yang mendekati hasil kontrol positif yaitu ekstrak etanol 96%. Hasil penurunan ekstrak etanol 96% sebesar 69,04 %, sedangkan nilai AUC paling kecil yang menunjukkan adanya aktivitas antidiabetes yaitu ekstrak etanol 96% dengan dosis 250 mg/KgBB sebesar 32695,31 dibandingkan dengan fraksi metanol 100% dengan dosis 250 mg/KgBB sebesar 33167,71. Simpulan dari hasil pengamatan kadar glukosa darah, ekstrak etanol 96% dosis 250 mg/KgBB memberikan potensi paling tinggi dalam menurunkan kadar glukosa darah. --- Kabau seed (Achidendron bubalinum (Jack) I.C Nielsen) is a plant that has antidiabetic properties. Empirically in North Sumatra, the kabau plant has been used as a traditional herbal antidiabetic medicine, besides the kabau seeds are used as a medicine for stomach aches and fever. This study was conducted to determine the antidiabetic activity of 96% ethanol extract and 100% methanol fraction of kabau seeds using glucose load method. This study used white male Swiss Webster mice divided into 5 treatment groups, namely normal control, positive control (glibenclamide 0.013mg / 20g mice BW), negative control (Na-CMC 0.5%), ethanol extract 96% ( 250mg / Kg BW for mice) and 100% methanol fraction (250mg / Kg BW for mice). The results showed a decrease in blood glucose levels relatively close to the positive control results, namely 96% ethanol extract. The result of the reduction in ethanol extract 96% was 69.04%, while the smallest AUC value that showed antidiabetic activity was 96% ethanol extract with a dose of 250 mg / KgBB of 32695.31 compared to the 100% methanol fraction at a dose of 250 mg / KgBW of 33167.71. The conclusion from the observation of blood glucose levels, 96% ethanol extract at a dose of 250 mg / KgBW provides the highest potential in reducing blood glucose levels.
