Abstract
Karakterisasi suatu zat aktif merupakan hal yang penting untuk diketahui sebelum melakukan formulasi sediaan farmasi untuk menghasilkan sediaan yang baik. Katekin merupakan salah satu zat aktif dari isolasi tanaman gambir yang memiliki efek farmakologi seperti antioksidan, antivirus, dan antibakteri. Tujuan penelitian ini adalah untuk mengetahui karakterisasi fisik isolat katekin dari tanaman gambir. Karakterisasi isolat katekin yang dianalisis adalah pengujian kelarutan, penentuan densitas, karakterisasi kristalografi menggunakan Powder X-ray Diffractometry (PXRD), karakterisasi morfologi permukaan kristal menggunakan Scanning Electron Microscopy (SEM), analisis sifat termal menggunakan Differential Scanning Calorimeter (DSC), dan pengukuran nilai zeta potensial menggunakan Zeta Potential Analyzer (ZPA). Hasil penelitian ini menunjukkan isolat katekin agak larut dalam etanol 96% dan gliserin, larut dalam PEG 400, mudah larut dalam propilenglikol dan sangat sukar larut dalam sorbitol. Nilai kerapatan sejati 1,299±0,0035 g/mL, indeks kompresibilitas 32,110±0,85%, rasio Hausner 1,472±0,018, berbentuk kristal dengan derajat kristalinitas 51,73%, memiliki permukaan morfologi yang kasar, Titik leleh 195,25 , titik dekomposisi 210 , dan nilai potensial zeta -13,167±0,288 mV. Berdasarkan hasil penelitian dapat disimpulkan bahwa isolat katekin memiliki kelarutan yang baik pada propilenglikol dan PEG 400, memiliki sifat alir dan stabilitas larutan yang kurang baik. --- Characterization of an active substance is an important thing to know before formulating pharmaceutical preparations to produce good preparations. Catechins are one of the active substances from the isolation of gambir plants that have pharmacological effects such as antioxidants, antivirals, and antibacterials. The purpose of this study was to determine the physical characterization of catechin isolates from gambir plants. The characterization of the catechin isolates analyzed were solubility testing, density determination, crystallography characterization using Powder X-ray Diffractometry (PXRD), crystal surface morphological characterization using Scanning Electron Microscopy (SEM), thermal properties analysis using Differential Scanning Calorimeter (DSC), and measurement of potential zeta values using Zeta Potential Analyzer (ZPA). The results of this study showed that catechin isolates are somewhat soluble in 96% ethanol and glycerin, soluble in PEG 400, easily soluble in propilenglikol and very difficult to dissolve in sorbitol. True density value of 1.299±0.0035 g/mL, compressibility index of 32.110±0.85%, Hausner ratio of 1.472±0.018, crystalline with a degree of crystallinity of 51.73%, has a rough morphological surface, Melting point of 195.25°C, decomposition point of 210°C, and potential zeta value of -13.167±0.288 mV. Based on the results of the study, it can be concluded that catechin isolates have good solubility in propilenglikol and PEG 400, have poor flow properties and solution stability
