Abstract

Kencur merupakan obat tradisional yang tidak hanya digunakan sebagai bumbu dapur tetapi dipercaya mampu mengobati berbagai penyakit. Banyak data penelitian tentang kencur yang membuktikan adanya aktivitas antibakteri. Salah satu kandungan senyawa aktif dalam ekstrak kencur yaitu isolat etil pmetoksisinamat (EPMS) yang termasuk senyawa metabolit sekunder golongan ester, yang diduga mempunyai aktivitas antibakteri. Tujuan penelitian ini untuk mengetahui aktivitas antibakteri dari isolat EPMS terhadap bakteri Gram negatif (Salmonella typhi ATCC 14028) dan bakteri Gram positif (Enterococcus faecalis ATCC 29212), dengan parameter uji konsentrasi hambat minimum (KHM) dan Konsentrasi Bunuh Minimum (KBM) dari isolat. Uji aktivitas antibakteri dilakukan dengan metode difusi cakram. Aktivitas antibakteri dilakukan dengan terbentuknya zona bening di sekitar kertas cakram yang disebut zona hambat. Penelitian ini menggunakan 5 perlakuan konsentrasi yaitu 100 ppm, 125 ppm, 150 ppm, 200 ppm dan 225 ppm, serta Azithromycin sebagai kontrol positif dan DMSO sebagai kontrol negatif. Diameter zona hambat yang menunjukkan aktivitas antibakteri Salmonella typhi oleh variasi konsentrasi isolat EPMS sebesar 10,20 mm (100 ppm); 10,33 mm (125 ppm); 10,50 mm (150 ppm); 10,63 mm (200 ppm) dan 10,93 mm (225 ppm) sedangkan diameter zona hambat yang menunjukkan aktivitas antibakteri Enterococcus faecalis sebesar 8,43 mm (100 ppm); 9,26 mm (125 ppm); 9,36 mm (150 ppm); 9,86 mm (200 ppm) dan 10,00 mm (225 ppm). Hasil KHM dari isolat EPMS yang telah dilakukan terhadap bakteri Salmonella typhi didapatkan pada konsentrasi 100 ppm dengan diameter zona hambat sebesar 10,20 mm sedangkan pada Enterococcus faecalis didapatkan pada konsentrasi 225 ppm dengan diameter zona hambat sebesar 10,00 mm. Dan pada hasil pengujian KBM menunjukkan bahwa isolat EPMS tidak mempunyai aktivitas membunuh bakteri Salmonella typhi maupun Enterococcus faecalis. --- Kencur is a traditional medicine that is not only used as a spice but is believed to be able to treat various diseases. A lot of research data about kencur proves the existence of anti-bacterial activity. One of the active compounds of the kencur extract is ethyl p-methoxycinnamate (EPMS) isolate which is a secondary metabolite compound of the phenol group, which is thought to have antibacterial activity. The purpose of this study was to determine the antibacterial activity of EPMS isolates against Gram-negative bacteria (Salmonella typhi ATCC 14028) and Gram-positive bacteria (Enterococcus faecalis ATCC 29212), with the test parameters of the optimum concentration of EPMS isolates in concentration minimum inhibition (MIC) and Minimum Bactericidal Concentration (MBC). The antibacterial activity test was carried out by the disc diffusion method. Antibacterial activity was carried out by the formation of a clear zone around the paper disc called the inhibition zone. This study used 5 concentration treatments, namely 100 ppm, 125 ppm, 150 ppm, 200 ppm, and 225 ppm, and Azithromycin as a positive control and DMSO as a negative control. The diameter of the inhibition zone showing the antibacterial activity of Salmonella typhi by the concentration variation of the EPMS isolate was 10.20 mm (100 ppm); 10.33 mm (125 ppm); 10.50 mm (150 ppm); 10.63 mm (200 ppm) and 10.93 mm (225 ppm) while the diameter of the inhibition zone showing the antibacterial activity of Enterococcus faecalis was 8.43 mm (100 ppm); 9.26 mm (125 ppm); 9.36 mm (150 ppm); 9.86 mm (200 ppm) and 10.00 mm (225 ppm). The MIC results from EPMS isolates that have been carried out on Salmonella typhi bacteria were obtained at a concentration of 100 ppm with an inhibition zone diameter of 10.20 mm while Enterococcus faecalis was obtained at a concentration of 225 ppm with an inhibition zone diameter of 10.00 mm. And the results of the MBC test showed that EPMS isolates did not have an activity to kill Salmonella typhi and Enterococcus faecalis bacteria.
