Abstract
Biji kopi didasarkan pada kualitas yang merupakan aspek utama. Beberapa faktor yang dapat mempengaruhi kualitas biji kopi sehingga menghasilkan biji kopi yang berkualitas rendah. Biji kopi berkualitas rendah sering disebut defect. Biji kopi defect atau cacat ini seperti biji pecah atau tidak utuh, kopong, menghitam, keriput. Namun biji kopi defect maupun non defect masih memiliki senyawa aktivitas antioksidan. Tujuan dari penelitian ini adalah menguji aktivitas antioksidan dengan metode DPPH (2,2-difenil-1-pikrihidrazil) dengan parameter analisis dalam penilaian IC50 dan memformulasikan sediaan serum yang mengandung ekstrak biji kopi arabika defect. Serum di optimasi basis dengan memvariasikan basis gelling agent natrosol dengan variasi konsentrasi 0,75%, 1%, dan 1,25%. Hasil penelitian ini menunjukkan ekstrak biji kopi arabika defect memiliki aktivitas antioksidan dengan nilai IC50 sebesar 56,62 ppm, pada sediaan serum memiliki aktivitas antioksidan nilai IC50 K1 sebesar 294 ppm dan pada K2 sebesar 282 ppm dan basis formulasi sediaan serum ekstrak biji kopi arabika defect yang baik menggunakan gelling agent natrosol dengan konsentrasi 0,75%. --- Coffee beans are based on quality, which is the central aspect. Several factors can affect the quality of coffee beans, resulting in low-quality coffee beans. Low-quality coffee beans are often called defects. Defective coffee beans or defects such as broken or incomplete, hollow, blackened, or wrinkled. However, both defect and non-defect coffee beans still have antioxidant activity compounds. This study aimed to test the antioxidant activity with the DPPH method (2,2-diphenyl-1-picrihydrazil) with analytical parameters in the IC50 assessment and formulate a serum preparation containing defective arabica coffee bean extract. The serum was optimized by varying the base gelling agent natrosol with varying concentrations of 0.75%, 1%, and 1.25%. The results of this study showed that the defective Arabica coffee bean extract had antioxidant activity with an IC50 value of 56.62 ppm. In serum preparations, the IC50 value of K1 was 294 ppm and at K2 was 282 ppm. Better use the gelling agent natrosol with a concentration of 0.75%.
