Abstract
Oleoresin merupakan produk samping dari proses ekstraksi rimpang kunyit yang memiliki kandungan kimia yang potensial untuk dikembangkan, salah satunya βturmeron. β-turmeron memiliki khasiat sebagai antioksidan, namun, belum banyak penelitian terkait dengan isolasi senyawa β-turmeron serta pengujian aktivitas antioksidannya. Isolasi senyawa β-turmeron dari oleoresin kunyit dilakukan dengan metode adsorpsi menggunakan silika gel dengan berbagai variasi perbandingan. βturmeron dianalisis menggunakan Spektrofotometri UV-Vis, Spektrofotometri IR, KLT dan GC-MS serta menguji aktivitas antioksidannya menggunakan metode DPPH. Dari hasil Spektrofotometri UV-Vis, eluat hasil adsorpsi dengan perbandingan 1:1 menunjukkan panjang gelombang maksimum yang konsisten yaitu pada 236 nm. Hasil analisa GC-MS menunjukkan isolat β-turmeron yang diperoleh dari proses isolasi memiliki kemurnian 52,02% dengan rendemen isolatnya sebesar 33,63%. Hasil identifikasi dengan KLT menunjukkan keberadaan senyawa yang diduga sebagai β-turmeron dengan nilai Rf 0,6. Spektrum IR menunjukkan adanya vibrasi stretching gugus C=C aromatik pada bilangan gelombang 2958,45 cm-1 dan 2924,21 cm-1 , menunjukkan adanya pola serapan C=O stretching gugus karbonil pada 1683,04 cm-1 dan 1445,65 cm-1 dan menunjukkan pola vibrasi C-H bending pada 1113,71 cm-1 dan 1036,13 cm-1 . Pada pengujian aktivitas antioksidan, diketahui bahwa isolat β-turmeron yang diuapkan di suhu ruang memiliki nilai IC50 paling besar yaitu 7,07 ppm yang dikategorikan ke dalam antioksidan kuat. --- Oleoresin is a by-product of the turmeric rhizome extraction process which has the potential to be developed chemical content, one of which is β-turmerone. βturmerone have properties as antioxidants, however, there have not been many studies related to the isolation of β-turmerone compounds as well as testing their antioxidant activity. Isolation of β-turmerone compounds from turmeric oleoresins is carried out by the adsorption method using silica gel with various comparison variations. β-turmerone were analyzed using UV-Vis Spectrophotometry, IR Spectrophotometry, TLC and GC-MS and tested their antioxidant activity using the DPPH method. The results of UV-Vis Spectrophotometry, the adsorption in a ratio of 1: 1 showed a consistent maximum wavelength at 236 nm. The results of the GCMS analysis showed that the β-turmerone isolate obtained from the isolation process had a purity of 52.02% with an isolate yield of 33.63%. The results of identification with TLC showed the presence of a compound suspected to be a βturmerone with an Rf value of 0.6. The IR spectrum shows the presence of stretching vibrations of the aromatic C=C group at wave numbers 2958.45 cm-1 and 2924.21 cm-1, indicates the presence of an absorption pattern of C=O stretching carbonyl groups at 1683.04 cm-1 and 1445.65 cm-1 and shows a pattern of C-H bending vibration at 1113.71 cm-1 and 1036.13 cm-1. In testing antioxidant activity, it was found that β-turmeron isolates evaporated at room temperature had the largest IC50 value of 7.07 ppm which was categorized into strong antioxidants.
