Abstract
Biosurfaktan merupakan senyawa permukaan aktif yang berasal dari mikroorganisme yang memiliki sifat biodegradable, tidak berbahaya dan menggunakan sumber yang dapat diperbaharui. Salah satu mikroorganisme yang menghasilkan biosurfaktan yaitu bakteri Bacillus cereus. Bakteri Bacillus cereus berpotensi sebagai penghasil biosurfaktan golongan lipopeptida. Telah dilakukan penyejajaran gen biosurfaktan jenis fengycin dengan persen identity sekitar 80- 81%, dan penyejajaran gen iturin dengan persen identity sekitar 90.31 %. Penelitian ini dilakukan untuk mengisolasi gen pengkode enzim fengycin dan iturin menggunakan metode Polymerase Chain Reaction (PCR) serta karakterisasi produk menggunakan metode Fourier Transform Infrared (FTIR) untuk mengetahui golongannya. Hasil spektra IR menunjukan bahwa biosurfaktan yang dihasilkan oleh bakteri Bacillus cereus termasuk kedalam golongan lipopeptida siklik. Hasil PCR menunjukan tidak terdapat fragmen DNA pada gen fengycin sedangkan pada gen iturin menunjukan adanya fragmen DNA berukuran < 10kb yang diduga sebagai genom. --- Biosurfactants are surface active compounds derived from microorganisms that have biodegradable properties, are harmless and use sources that can be developed. One of the microorganisms that produce biosurfactants is the bacterium Bacillus cereus. Bacillus cereus bacteria may be a producer of lipopeptide biosurfactants. The alignment of the fengycin type of biosurfactant gene has been carried out with a percent identity of about 80-81%, and the alignment of the iturin gene with a percent identity of about 90.31%. This research was conducted to isolate the fengycin and iturin biosurfactant genes using the Polymerase Chain Reaction (PCR) method and product characterization using the Fourier Transform Infrared (FTIR) method to determine the group. The results of the IR spectra showed that the biosurfactants produced by Bacillus cereus were included in the cyclic lipopeptide group. The PCR results showed that there were no DNA fragments in the fengycin gene while the iturin gene showed the presence of < 10 kb DNA fragments that were forgotten as genomes.
