Abstract

Kurkumin merupakan salah satu senyawa kurkuminoid selain desmetoksikurkumin dan bisdemetoksikurkumin yang berasal dari rimpang tanaman kunyit (Curcuma Longa Linn). Dalam bidang pengobatan kurkumin telah terbukti memiliki aktivitas farmakologis yang beragam, seperti antiinflamasi, antioksidan, antivirus, antimalaria, antiparasit, dan antimikroba. Kelarutan merupakan salah satu parameter penting dalam proses isolasi, ekstraksi, pemurnian, rekristalisasi dan preformulasi suatu obat pada bidang industri. Penelitian ini bertujuan untuk mengetahui kelarutan kurkumin pada pelarut asetonitril, air, kloroform dan metanol dengan metode shake-flask saturation. Kadar kurkumin yang terlarut dalam masing-masing pelarut ditentukan dengan spektrofotometri dan gravimetri. Hasil uji kelarutan menunjukkan bahwa kurkumin memiliki kelarutan paling baik pada asetonitril dan paling buruk pada air dengan urutan kelarutan kurkumin dalam empat pelarut organik dari yang paling larut adalah asetonitril > metanol > kloroform > air. Kadar kurkumin terlarut berdasarkan metode gravimetri adalah 13,4575 mg/mL pada pelarut asetronitril, 11,97 mg/mL pada metanol, 7,665 mg/mL pada kloroform, 1,4075 mg/mL pada air, sedangkan berdasarkan metode spektrofotometri kadar kurkumin terlarut adalah 7,4215 mg/mL pada asetonitril, 5,5683 mg/mL pada metanol, 3,5441 mg/mL pada kloroform, dan 0,1657 mg/mL pada air. Data penelitian ini dapat dimanfaatkan untuk pengembangan sediaan obat berbahan dasar kurkumin dari proses isolasi hingga formulasi. --- Curcumin is one of the curcuminoid compounds besides desmethoxycurcumin and bisdemethoxycurcumin derived from the rhizome of the turmeric plant (Curcuma Longa Linn). In the field of medicine, curcumin has been shown to have diverse pharmacological activities, such as anti-inflammatory, antioxidant, antiviral, antimalarial, antiparasitic, and antimicrobial. Solubility is one of the important parameters in the process of isolation, extraction, purification, recrystallization and preformulation of a drug in the industrial field. This study aims to determine the solubility of curcumin in acetonitrile, water, chloroform and methanol solvents using the shake-flask saturation method. The concentration of curcumin dissolved in each solvent was determined by spectrophotometry and gravimetry. The results of the solubility test showed that curcumin had the best solubility in acetonitrile and the worst in water with the order of solubility of curcumin in four organic solvents from most soluble to acetonitrile > methanol > chloroform > water. The dissolved curcumin level based on the gravimetric method was 13.4575 mg/mL in acetronitrile, 11.97 mg/mL in methanol, 7.665 mg/mL in chloroform, 1.4075 mg/mL in water, while based on the spectrophotometric method, the dissolved curcumin content was 7.4215 mg/mL in acetonitrile, 5.5683 mg/mL in methanol, 3.5441 mg/mL in chloroform, and 0.1657 mg/mL in water. This research data can be used for the development of curcumin-based drug preparations from the isolation process to the formulation.
