Abstract
Antioksidan merupakan senyawa yang menghambat atau menunda reaksi oksidasi molekul dengan cara menghambat proses inisiasi atau propagasi reaksi oksidasi berantai. Penelitian bertujuan untuk mengetahui potensi antioksidan yang paling kuat dari ekstrak biji kabau (Archidendron bubalinum (Jack.) I.C Nieslen.) yang di fraksi menggunakan eluen N-heksana, etil aseatat dan aquadest. Sebelum pengujian aktivitas antioksidan dilakukan kromatografi lapis tipis (KLT). Pengujian aktivitas antioksidan dari ketiga fraksi dan ekstrak biji kabau ini yaitu dilakukan dengan metode DPPH yang kemudian diamati dengan spektrofotometer UV-Vis pada panjang gelombang maksimum 513-514 nm. Analisis data dilakukan dengan menghitung persen inhibisi yang kemudian dibuat persamaan regresi linear menggunakan Microsoft Excel. Hasil yang diperoleh bahwa nilai IC60 ekstrak kabau yaitu µg/mL, nilai IC60 dari fraksi N-heksana yaitu 18,99 µg/mL, nilai IC60 dari fraksi etil aseatat yaitu 7,583 µg/mL dan nilai IC60 dari fraksi aquadest yaitu 21,51µg/mL serta nilai IC60 dari ekstrak kabau 28,65. Dari hasil tersebut potensi antioksidan paling besar yaitu pada fraksi etil asetat, semakin kecil nilai inhibitor consentration maka semakin besar aktivitas antioksidan. --- Antioxidants are compounds that inhibit or delay molecular oxidation reactions by inhibiting the initiation or propagation of chain oxidation reactions. The aim of the study was to determine the strongest antioxidant potential of kabau seed extract (Archidendron bubalinum (Jack.) I.C Nieslen.) which was fractionated using N-hexane, ethyl acetate and aquadest as eluents. Before testing the antioxidant activity, thin layer chromatography (TLC) was performed. Testing the antioxidant activity of the three fractions and extracts of kabau seeds was carried out using the DPPH method which was then observed with a UV-Vis spectrophotometer at a maximum wavelength of 513-514 nm. Data analysis was carried out by calculating the percentage of inhibition which was then made a linear regression equation using Microsoft Excel. The results obtained that the IC60 value of the kabau extract was g/mL, the IC60 value of the N-hexane fraction was 18.99 g/mL, the IC60 value of the ethyl acetate fraction was 7.583 g/mL and the IC60 value of the aquadest fraction was 21.51µg/ mL and the IC60 value of kabau extract 28.65. From these results, the greatest antioxidant potential is in the ethyl acetate fraction, the smaller the inhibitor concentration value, the greater the antioxidant activity.
