Abstract

Infeksi adalah penyakit yang ditularkan dari satu orang ke orang lain atau dari hewan ke manusia yang disebabkan oleh mikroorganisme seperti bakteri, virus, jamur, dan protozoa. Piperin merupakan senyawa metabolit sekunder yang diperoleh dari buah lada (Piper albi L). Piperin adalah senyawa alkaloid yang terdapat dalam tanaman famili Piperaceae, salah satu efek yang dimiliki piperin adalah sebagai anti-mikroba. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri dari isolat piperin terhadap bakteri Salmonella thypi ATCC 14028, dan Enterococcus faecalis ATCC 29212, dan juga untuk mengetahui berapa konsentrasi optimum dalam menghambat (KHM) atau membunuh (KBM) terhadap bakteri Salmonella thypi ATCC 14028, dan Enterococcus faecalis ATCC 29212. Metode uji aktivitas antibakteri yang digunakan dalam penelitian ini yaitu difusi cakram dengan variasi konsentrasi 100 ppm, 125 ppm, 150 ppm, 200 pm, dan 225 ppm. Aktivitas antibakteri dilihat dari terbentuknya zona hambat. Hasil uji aktivitas antibakteri dari kelima konsentrasi larutan uji menunjukkan diameter zona hambat pada bakteri Salmonella thypi ada pada konsentrasi 150 ppm sebesar 10,07 mm, sedangkan bakteri Enterococcus faecalis ada pada konsentrasi 200 ppm sebesar 10,05 mm. Konsentrasi Bunuh Minimum (KBM) pada isolat piperin menunjukkan bahwa isolat tersebut tidak mampu membunuh bakteri Salmonella thypi dan Enterococcus faecalis. Isolat piperin ini mempunyai kekuatan antibakteri kriteria sedang, dengan diameter zona hambat rata-rata 10-14 mm. --- Infection is a disease that is transmitted from one person to another or from animals to humans caused by microorganisms such as bacteria, viruses, rickettsiae, fungi, and protozoa. Piperine is a secondary metabolite compound obtained from pepper fruit (Piper albi L). Piperine is an alkaloid compound found in the Piperaceae family of plants, one of the effects of piperine is as an anticancer and anti-microbial. This study aims to determine the antibacterial activity of piperine isolates against Salmonella thypi ATCC 14028 and Enterococcus faecalis ATCC 29212 bacteria and also to determine the optimum concentration in inhibiting (MIC) or killing (MBC) against Salmonella thypi ATCC 14028 and Enterococcus faecalis ATCC 29212 bacteria. The antibacterial activity test method used in this study namely disc diffusion with various concentrations of 100 ppm, 125 ppm, 150 ppm, 200 pm, and 225 ppm. Antibacterial activity was seen from the formation of the inhibition zone. The results of the antibacterial activity test of the five concentrations of the test solution showed that the diameter of the inhibition zone for Salmonella typhi bacteria was at a concentration of 150 ppm at 10,07 mm, while Enterococcus faecalis was at a concentration of 200 ppm at 10,05 mm. Minimum Kill Concentration (KBM) on piperine isolates showed that the isolates were unable to kill Salmonella typhi and Enterococcus faecalis bacteria. This piperine isolate had moderate antibacterial strength, with an average inhibition zone diameter of 10-14 mm.
