Abstract
Biji kabau (Archidendron bubalinum (Jack) I.C Nielsen) memiliki kandungan kimia seperti flavonoid dan polifenol yang berpotensi sebagai antioksidan yang dapat berfungsi menangkal radikal bebas dan dapat menyebabkan berbagai macam penyakit. Penelitian ini bertujuan untuk mengetahui dan membandingkan aktivitas antioksidan dari fraksi etil asetat, etil asetat : metanol (1:1) dan metanol dari ekstrak biji kabau dengan metode penangkapan radikal 2,2-difenil-1pikrilhidrazil (DPPH). Biji kabau diekstraksi mengunakan pelarut etanol 96% dengan menggunakan metode soxhletasi, kemudian difraksinasi dengan metode cair padat menggunakan pelarut etil asetat, etil asetat : metanol (1:1), dan metanol. Fraksi yang diperoleh kemudian dianalisis aktivitas antioksidannya dengan metode DPPH. Pengukuran dilakukan secara spektrofotometri pada panjang gelombang 514 nm. Aktivitas antioksidan dinyatakan dengan parameter IC50. Hasil uji aktivitas antioksidan menunjukkan bahwa semua konsentrasi memiliki kemampuan menangkap radikal bebas DPPH dengan nilai IC50 untuk fraksi etil asetat, etil asetat : metanol (1:1), metanol, dan vitamin C berturut turut adalah 85,96 ppm; 98,08 ppm; 131,93 ppm; dan 29,39 ppm. --- Kabau seeds (Archidendron bubalinum (Jack) I.C Nielsen) contain chemicals such as flavonoids and polyphenols that have the potential as antioxidants that can function to ward off free radicals and can cause various diseases. This study aims to determine and compare the antioxidant activity of the fractions of ethyl acetate, ethyl acetate: methanol (1:1) and methanol from kabau seed extract using the 2,2-diphenyl1picrylhydrazyl (DPPH) radical scavenging method. Kabau seeds were extracted using 96% ethanol as solvent using the soxhletation method, then fractionated by liquid solid method using ethyl acetate, ethyl acetate: methanol (1:1), and methanol as solvents. The obtained fraction was then analyzed for its antioxidant activity using the DPPH method. Measurements were made spectrophotometrically at a wavelength of 514 nm. Antioxidant activity is expressed by parameter IC50. The results of the antioxidant activity test showed that all concentrations had the ability to scavenge DPPH free radicals with IC50 values for the ethyl acetate fraction, ethyl acetate: methanol (1:1), methanol, and vitamin C 85.96 ppm, respectively; 98.08 ppm; 131.93 ppm; and 29.39 ppm.
