Abstract

Etil p-metoksisinamat (EPMS) larut pada sebagian pelarut polar dan nonpolar sehingga memiliki kelarutan dan disolusi yang rendah. Penelitian EPMS masih terbatas pada peningkatan kelarutan dan disolusi EPMS sehingga data penelitian secara in vivo menggunakan plasma darah hewan uji masih terbatas. Penelitian metode analisis EPMS pada plasma darah hewan uji menggunakan Kromatografi Cair Kinerja Tinggi (KCKT) belum tervalidasi. Penelitian ini bertujuan untuk melakukan validasi metode analisis EPMS dalam plasma darah menggunakan KCKT. Plasma darah yang digunakan merupakan plasma darah tikus putih jantan galur Wistar. EPMS ditambahkan ke dalam plasma darah dan dianalisis menggunakan KCKT. Parameter validasi yang digunakan adalah uji kesesuaian sistem, linearitas, akurasi, presisi, LOD, dan LOQ. Hasil penelitian menunjukkan fase gerak yang optimum untuk menganalisis EPMS di dalam plasma adalah metanol asam fosfat dengan perbandingan 70:30. Berdasarkan hasil validasi didapatkan hasil regresi linier 0,779. Pada uji akurasi didapatkan rentang nilai rata-rata persen perolehan kembali sebesar 71.55% 168,04%. Pada uji presisi didapatkan rentang nilai rata-rata persen RSD sebesar 1.02%-3.14%. Nilai LOD sebesar 5,8383 µg/ml. dan nilai LOQ sebesar 19,4611 µg/mL. Berdasarkan penelitian ini, dapat disimpulkan bahwa analisis EPMS dalam plasma darah menggunakan KCKT dapat dianalisis menggunakan fase gerak metanolasam fosfat dengan perbandingan 70:30. --- Ethyl p-methoxycinnamate (EPAMC) is soluble in most polar and non-polar solvents so that it has low solubility and dissolution. Research on EPMC is still limited to increasing the solubility and dissolution of EPMC. In vivo research data using the blood plasma of test animals is still limited. Research on the EPMC analysis method on blood plasma of text animals using High Performance Liquid Chromatography (HPLC) has not been validated. This study aims to validate the EPMC analysis method in blood plasma using HPLC. The blood plasma used was the blood plasma of male white rats of the Wistar strain. EPMC was added to blood plasma and analyzed using HPLC. The validation parameters used are system suitability test, linearity, accuracy, precision, LOD, and LOQ. The results showed that the optimum mobile phase for analyzing EPMC in plasma was methanol:phosphoric acid in a ratio of 70:30. Based on the validation results obtained linear regression results of 0.779. In the accuracy test, the average value range of % recovery is 71.55%-168.04%. In the precision test, the average value range of % RSD is 1.02%-3.14% The LOD value is 5,8383 g/ml, and the LOQ value is 19.4611 g/ml.
