Abstract
Andrografolid merupakan komponen bioaktif paling utama yang terkandung dalam tanaman sambiloto (Andrographis paniculata Burm. F. Nem). Senyaws andrografolid merupakan diterpenoid lakton yang memiliki cincin lakton pada strukturnya. Penelitian ini bertujuan untuk memurnikan andrografolid menggunakan tiga jenis adsorben yaitu moleculary imprinted polymer (MIP), moleculary imprinted polymer berbasis silika mikrosfer (MIPS), Moleculary imprinted polymer berbasis silika gel (MIP SG). Crude andrografolid yang dimunikan didapatkas dari ekstrak daun sambiloto. Pada peselitian ini dilakukan optimasi untuk menentukan waktu kontak dan berat adsorben optimal serta splikasinya untuk pemurnian andrografolid. Hasil optimasi menunjukan bahwa waktu kontak optimal MIP & MIPs terjadi pada waktu 120 menit, untuk MIP SG terjadi pada waktu 60 menit. Berat adsorben optimal untuk ketiga adsorben yains 10 mg. Setelah diperoleh waktu dan berat optimal adsorben, ketiga adsorben diaplikasikan untuk pemmarnian erade andrografolid yang mana kemurnian androgrfaolid ditentukan dengan membandingkan perses ares crude dengan persen ares crude setelah melalui tahap pemurnian. Berdasarkan hasil kromatogram KCKT dapat diketahui bahwa MIP dapat meningkatkan persen ares crude dari 0,79% menjadi 15,34%, MIPs dapat meningkatkan persen ares crude dari 0,79% menjadi 17,64% dan MIP SG dapat menigkatkan persen area crude dari 0,79% menjadi 20,57% --- Andrographolide is the most important bioactive component contained in bitter plant (Andrographis paniculata Burm. F. Ness). Andrographolide is a lactone diterpenoid which has a lactone ring in its structure. This study aimed to purify andrographolide using three types of adsorbents, namely moleculary imprinted polymer (MIP), moleculary imprinted polymer based on silica microspheres (MIPs), and Molecular imprinted polymer based on silica gel (MIP SG). Crude purified andrographolide was obtained from bitter leaf extract. In this study, optimtration was carried out to determine the optimal contact time and weight of the adsorbent and its application for andrographolide purification. The optimization results showed that the optimal contact time for MIP & MIP occured at 120 minutes, for MIP SG it occurs at 60 minutes. The optimal adsorbent weight for the three adsorbents were 10 mg. After obtaining the optimal time and weight of the adsorbent, the three adsorbents were applied for the purification of crude andrographolide in which the purity of the andrographolide was determined by comparing the percent ofarea crude with the percent ofarea crude after going through the purification stage, Based on the results of the HPLC chromatogram, it can be seen that MIP can increase the percent ofaren crude from 0.79% to 15.34%, MIPs can increase the percent ofarea crude from 0.79% to 17.64% and MIP SG can increase the percentares crude offrom 0.79% to 20.5756 M
