Abstract
Brazilin termasuk kedalam kelas IV pada Sistem Klasifikasi Biofarmasi karena memiliki sifat kelarutan dan permeabilitas yang buruk. Untuk mengatasi masalah tersebut, perlu dilakukan upaya untuk meningkatkan kelarutan brazilin, salah satunya yaitu dengan teknik dispersi padat amorf. Penelitian ini bertujuan untuk mengetahui pengaruh PVP terhadap karakterisasi yang dapat memprediksi peningkatan kelarutan pada dispersi padat. Dispersi padat amorf dibuat menggunakan metode pelarutan (solvent evaporation) dengan perbandingan bobot 1:1 (F1), 1:3 (F2), dan 1:5 (F3). Karakterisasi dilakukan menggunakan Powder XRay Diffraction (PXRD), Differential Scanning Calorimetry (DSC), dan Fourier Transform Infrared Spectroscopy (FTIR). Hasil karakteristik dengan PXRD menunjukkan penurunan kristalinitas yang signifikan menjadi lebih amorf pada ketiga formula dibandingkan brazilin murni. Analisis DSC mengkonfirmasi adanya interaksi antara brazilin dengan PVP K-30 yang mengindikasikan perubahan sifat termal kearah yang lebih amorf. Hasil analisis FTIR menunjukkan bahwa terjadi interaksi secara kimiawi antara isolat brazilin dan PVP K-30. ---- Brazilin is included in class IV in the Biopharmaceutical Classification System because it has poor solubility and permeability properties. To overcome this problem, efforts need to be made to increase the solubility of brazilin, one of which is using the amorphous solid dispersion technique. This research aims to determine the effect of PVP on characterization which can predict increased solubility in solid dispersions. Amorphous solid dispersions were made using the solvent evaporation method with weight ratios of 1:1 (F1), 1:3 (F2), and 1:5 (F3). Characterization was carried out using Powder X-Ray Diffraction (PXRD), Differential Scanning Calorimetry (DSC), and Fourier Transform Infrared Spectroscopy (FTIR). Characteristic results with PXRD showed a significant decrease in crystallinity to become more amorphous in all three formulas compared to pure brazilin. DSC analysis confirmed the interaction between brazilin and PVP K-30 which indicated a change in thermal properties towards a more amorphous direction. The results of FTIR analysis showed that there was a chemical interaction between the brazilin isolate and PVP K-30.
