Abstract

Kurkumin, senyawa aktif yang ditemukan dalam kunyit, memiliki efek antiinflamasi dan penangkal radikal bebas. Potensinya untuk dikembangkan menjadi sediaan tablet terhambat oleh bioavailabilitas yang rendah. Hal ini mendorong pengembangan Sediaan Oral Pelepasan Terkendali (SOPT) solid untuk mengoptimalkan efektivitasnya. Sebagai bagian dari proses preformulasi, diperlukan pengujian inkompatibilitas kurkumin terhadap eksipien yang digunakan dalam SOPT solid, seperti aerosil, kalsium stearat, dan natrium klorida. Penelitian ini bertujuan untuk menganalisis potensi inkompatibilitas antara kurkumin dan eksipien yang digunakan dalam SOPT solid, yaitu aerosil, kalsium stearat, dan natrium klorida. Metode penelitian meliputi analisis organoleptik, morfologi, homogenitas, kristalinitas, sifat termal, Loss on Dry (LoD), dan Fourier Transform Infrared (FTIR). Preparasi sampel dilakukan dengan mencampurkan kurkumin dengan eksipien dengan perbandingan 1:1 pada kecepatan 60 rpm selama 15 menit. Hasil penelitian menunjukkan kurkumin dengan aerosil, kalsium stearat, dan natrium klorida memiliki homogenitas yang rendah. Difraktogram, termogram dan spektrum campuran kurkumin dengan aerosil, kalsium stearat, dan natrium klorida menunjukkan bahwa sampel mempertahankan struktur dan bentuknya masingmasing. Hasil LoD dari masing-masing sampel campuran memenuhi persyaratan. Berdasarkan penelitian ini, dapat disimpulkan bahwa kurkumin dengan NaCl, aerosil, dan kalsium stearat diduga memiliki potensi yang rendah untuk mengalami inkompatibilitas. Penelitian lanjutan perlu dilakukan untuk memastikan potensi ini. ----- Curcumin, the active compound found in turmeric, has anti-inflammatory and antioxidant effects. Its potential for development into tablet formulations is hindered by low bioavailability. This challenge has prompted the development of solid Controlled Release Oral Formulations (CROF) to optimize its effectiveness. As part of the preformulation process, it is necessary to test the incompatibility of curcumin with excipients used in CROF solid forms, such as aerosil, calcium stearate, and sodium chloride. This study aimed to analyze the potential incompatibility between curcumin and the excipients used in solid Controlled Release Oral Formulations (CROF), namely aerosil, calcium stearate, and sodium chloride. Sample preparation was carried out by mixing curcumin with excipients in a ratio of 1:1 at 60 rpm for 15 minutes. The results showed low homogeneity of curcumin with aerosil, calcium stearate, and sodium chloride. Diffractograms, thermograms and spectrum of curcumin mixtures with aerosil, calcium stearate, and sodium chloride indicated that the samples maintained their respective structures and forms. LoD results from each sample mixture met the requirements. Based on this research, it was concluded that curcumin with NaCl, aerosil, and calcium stearate was suspected to have a low potential for incompatibility. Further research is needed to confirm this potential.
